摘  要:目的应用离体高分辨魔角旋转磁共振波谱(high resolution magic angle spinning nuclear magneticresonance,HR-MAS1HNMR)技术检测动脉粥样硬化兔锁骨下动脉盗血模型海马区的神经代谢变化。方法 24只雄性新西兰大白兔随机分为普通饮食对照组、高脂饮食组和高脂饮食结扎组,每组8只。采用右侧锁骨下动脉结扎法建立动脉粥样硬化兔锁骨下动脉盗血模型。用离体高分辨魔角旋转磁共振波谱技术检测兔海马组织代谢产物N-乙酰-天冬氨酸复合物(NAA)、胆碱(Cho)、肌酸/磷酸肌酸(Cre)、乳酸(Lac)、γ-氨基丁酸(GABA)、谷氨酸(Glu)等共振峰的变化。结果与普通饮食对照组及高脂饮食组比较,模型组兔海马区NAA、GABA明显降低(P<0.05),Glu显著升高(P<0.05),其他代谢产物变化不明显。普通饮食对照组与高脂饮食组代谢产物比较无明显变化。结论动脉粥样硬化兔锁骨下动脉盗血模型对海马神经元代谢影响显著,神经元功能受损,为动脉粥样硬化性脑缺血的研究提供了可靠的实验理论依据。Objective To study the changes of metabolites in hippocampus CA1 sections of rabbit subclavian steal model by high resolution magic angle spinning nuclear magnetic resonance(HR-MAS1HNMR).Methods Twenty-four male New Zealand white rabbits were randomly assigned to 3 groups with 8 animals in each group: rabbits were fed with normal diet(normal control) or high-fat diet(high-fat control),and fed with high-fat diet plus surgical ligation of right subclavian artery(model group).The metabolites changes in hippocampus were analyzed by HRMAS1HNMR,including N-acetylaspartate(NAA),choline(Cho),creatine(Cre),Lactic acid(Lac),γ-Amino butyric acid(GABA) and glutamate(Glu).Results Compared with the normal control group and high-fat control group,the concentrations of NAA and GABA in the model group decreased significantly,and that of Glu was increased;while the concentrations of other metabolites were not significantly changed.Conclusion The subclavian steal model of atherosclerotic rabbits demonstrates that the metabolism and function of hippocampal neurons are significantly disturbed,which provides the experimental evidence for the study of ischemic atherosclerotic diseases.
