摘 要:目的观察宣肺中药(加味桔梗汤)对便秘小鼠肠道传输功能和肺、肠组织速激肽A(NKA)、血管活性肠肽(VIP)含量的影响,从神经肽角度探讨加味桔梗汤防治慢性传输型便秘的现代生物学机制,"肺合大肠"脏腑相关联络机制。方法 40只小鼠随机分为正常组、模型组、模型给药组及正常给药组,采用自身粪便与复方地芬诺酯(2 mg/kg)联合灌胃法建立慢传输型便秘模型。模型给药组及正常给药组以加味桔梗汤(12 g/kg)灌胃,正常组、模型组以蒸馏水灌胃,连续7 d。观察小鼠排便情况,测定肺、肠组织中NKA和VIP含量。结果与正常组比较,模型组小鼠首粒黑便时间延长,4 h黑便粒数与重量明显减少,肺肠组织中NKA含量明显升高,VIP含量明显降低;与模型组比较,模型给药组首粒黑便时间缩短,4 h排便粒数与重量增加,肺肠组织中NKA含量明显降低,肺组织中VIP含量明显升高。结论加味桔梗汤可改善便秘小鼠肠道传输功能,其机制可能与调节肺、肠组织中神经肽NKA、VIP的含量相关。Objective To observe the influences of Chinese medicinals with actions of dispersing lung ( modified Jiegeng Tang) on investine transmission function and content of neurokinin A ( NKA ) and vasoactive intestinal peptide (VIP) in lung and intestine in mice with slow transit constipation (STC), and discuss the connecting mechanism of lung associating with large intestine from the angle of neuropeptides. Methods Mice (n = 40) were randomly divided into normal group, model group, model treatment group and normal treatment group. STC model was established with the intragastrical method combining self-feces and compound diphenoxylate. Model treatment group and normal treatment group were given modified Jiegeng Tang intragastrically for 7 days, and normal group and model group was given distilled water intragastrically for 7 days. The defecation of mice was observed and content of NKA and VIP were detected in lung and intestine. Results Compared with normal group, in model group the time of the first melena was prolonged, number and weight of 4-hour melena decreased significantly, and NKA increased significantly and VIP decreased significantly in lung and intestine. Compared with model group, in model treatment group the time of the first melena was shortened, number and weight of 4-hour melena increased, and NKA decreased significantly in lung and intestine and VIP increased significantlyin lung. Conclusion Modified Jiegeng Tang mice, and the mechanism may be related to and intestine. can improve the intestine transmission function in STC regulation of the content of NKA and VIP in lung
