摘  要:目的:探讨雌激素受体在大鼠肝脏缺血再灌注损伤中的表达和意义。方法:采用Pringle’s法制作肝脏缺血再灌注损伤模型。选取健康清洁级雄性SD大鼠90只,随机分成假手术组(PO组)、肝缺血再灌注组(IR组)和雌激素预处理+肝缺血再灌注组(IRE组)3组,每组各30只。各组分别取肝缺血前(T0)、再灌注1h(T1)、再灌注3h(T3)、再灌注6h(T6)、再灌注24h(T24)共5个时间点的血液标本,检测血清谷丙转氨酶(ALT)浓度,HE染色观察肝脏组织形态学变化,ELISA法测定血清肿瘤坏死因子-α(TNF-α)水平,免疫组化方法检测肝脏组织核转录因子(NF-κB)和雌激素受体(ER)的表达情况。结果:IR组、IRE组与PO组比较,血清ALT、TNF-α水平升高(P<0.01),但IRE组明显低于IR组(P<0.05);肝脏组织形态学IR组和IRE组可见肝脏组织损伤,但IRE组明显轻于IR组;IR组、IRE组与PO组比较,肝组织ER表达升高(P<0.01),但IRE组明显高于IR组(P<0.05);IR组、IRE组与PO组比较,肝组织NF-κB表达明显升高(P<0.01),但IRE组明显低于IR组(P<0.05)。结论:雌激素预处理能刺激雌激素受体表达,进而降低TNF-α水平,抑制NF-κB表达来减轻肝脏缺血再灌注损伤。Objective:To investigate the expressions of estrogen receptor in hepatic ischemia reperfusion injury in rats. Methods:90 healthy male SD rats were divided into 3 groups randomly:pseudo-operation group(PO), hepatic ischemia reperfusion group(IR), estrogen pretreatment plus hepatic ischemia reperfusion group(IRE). Serum ALT and TNF-α levels , NF-κB and estrogen receptor (ER) expression were measured in time of pre-ischemia(T0 ), reperfusion 1 (T1), 3 (T3), 6 (T6) and 24 hours(T24 ) respectively. Results : NF-κB, ALT and TNF-α levels in IR and IRE group were increased compared to PO,the values of group IRE were lower than those in group IR. The ER ex- pression of group IR and IRE were increased significantly compared to group PO, but ER expression of IRE group was higher than those in IR group significantly. The liver damage of group IR and IRE were more severe than those in group PO,but the group IR were more severe than those in group IRE. Conclusion:Estrogen pretreatment can sig- nificantly increase the estrogen receptor expression , alleviate hepatic ischemia reperfusion injury by decreasing NF- κB and TNF-α level in IR rat medel.
