摘 要:目的:比较黄连不同炮制品中5种生物碱的含量差异。方法:采用高效液相色谱法测定含量,色谱柱为Ultimate XB-C_(18)色谱柱(250 mm×4.6 mm,0.45μm);流动相:乙腈-(磷酸-三乙胺-水,1:2:100)(26:74);流速:1.0 rnl·min^(-1);检测波长:265 nm;柱温:30℃。结果:黄连各炮制品中的盐酸药根碱含量为0.36%~0.41%,其中姜黄连>萸黄连>酒黄连>生黄连:盐酸表小檗碱含量为0.91%~1.06%,其中萸黄连>姜黄连>生黄连>酒黄连;盐酸黄连碱含量为1.81%~2.09%,其中生黄连>酒黄连>萸黄连>姜黄连;盐酸巴马汀含量为1.42%~1.72%,其中萸黄连>生黄连>酒黄连>姜黄连;盐酸小檗碱含量为6.74%~7.12%,其中姜黄连>萸黄连>生黄连>酒黄连。结论:不同炮制方法对黄连5种生物碱含量均有不同的影响,为进一步探讨黄连的炮制机制提供试验基础。Objective:To compare the content difference of five alkaloids in rhizoma coptidis with different processing methods. Method:An HPLC was used to determine the alkaloids with an Ultimate XB-C_(18)(250 mm×4.6 mm,0.45μm) column.The mobile phase was acetonitrile-(phosphoric acid-triethylamine-water,1:2:100)(26:74),the flow rate was 1 ml? min^(-1),the detection wavelength was 265 nm,and the column temperature was 301.Result:In the different processed products of rhizoma coptis,the content of jatrorrhizine hydrochloride was 0.36%-0.41%with the order of the product with ginger the product with fructus evodiae the product with yellow wine the crude.The content of epiberberine hydrochloride was 0.91%-1.06%with the order of the product with fructus evodiae the product with ginger the crude the product with yellow wine.The content of coptisine hydrochloride was 1.81%-2.09%with the order of the crude the product with yellow wine the product with fructus evodiae the product with ginger. The content of palmatine hydrochloride was 1.42%-1.72%with the order of the product with fructus evodiae the crude the product with yellow wine the product with ginger.The content of berberine hydrochloride was 6.74%-7.12%with the order of the product with ginger the product with fructus evodiae the crude the product with yellow wine.Conclusion:Different processing methods show different effects on the contents of the 5 alkaloids in rhizoma coptis,and the study can provide the experimental basis for the further study on the processing mechanism of rhizoma coptis.
