摘  要:目的:建立同时测定复方氨基酸(15)双肽(2)注射液中焦谷氨酸、环(甘氨酰-谷氨酰胺)和环(甘氨酰-酪氨酸)等杂质含量的HPLC法。方法:使用Agela venusil ASB C_(18)色谱柱(250 mm×4.6 mm,5μm),流动相为0.01 mol·L^(-1)磷酸氢二铵溶液(用磷酸调节pH至1.6)-甲醇(90:10),流速为1.0 ml·min^(-1),检测波长为205 nm;柱温为35℃;进样量为50μl。结果:焦谷氨酸、环(甘氨酰-谷氨酰胺)和环(甘氨酰-酪氨酸)浓度分别在29.47~589.37(r=0.999 7),47.94~958.82(r=0.999 8)和4.84~96.9 mg·L^(-1)(r=0.999 8)范围内线性关系良好,r值分别为0.999 7,0.999 8,0.999 8。检出限分别为1.47,2.49和0.29 ng,平均回收率分别为99.6%,99.9%和100.6%。结论:本方法简便、准确,可用于复方氨基酸(15)双肽(2)注射液中焦谷氨酸、环(甘氨酰-谷氨酰胺)和环(甘氨酰-酪氨酸)含量的快速分析。Objective:To establish a method for the determination of pyroglutamic acid,cycol(Gly-Gln) and cycol(Gly-L-Tyr) in compound amino acids(15) and dipeptides(2) injections by HPLC.Method:An Agela venusil ASB -C_(18)(250 mm×4.6 mm,5μm) column was applied.The mobile phase was 0.01 mol·L^-1 ammonium dibasic phosphate solution(adjusting pH to 1.6 with phosphoric acid)-methanol(90:10) at a flow rate of 1.0 ml · min^-1,the detection wavelength was 205 nm,the column temperature was 35℃, and the injection volume was 50μl.Result:There was good linearity within the range of 29.47-589.37 mg · L^-1(r = 0.999 7) for pyroglutamic acid,47.94-958.82 mg · L^-1(r = 0.999 8) for cyclo(Gly-Gln) and 4.84-96.9 mg ? L^-1(r=0.999 8) for cyclo(GlyL -Tyr),respectively.The limit of detection of pyroglutamic acid,cycol(Gly-Gln) and cycol(Gly-L-Tyr) was 1.47,2.49 and 0.29 ng,and the recovery was 99.6%,99.9%and 100.6%,respectively.Conclusion:The method is simple and accurate,and can be used in the fast analysis of the preparation.
