摘  要:目的观察癌-睾丸抗原SSX基因在肝癌组织和肝癌细胞株中mRNA水平的表达情况,探究该基因家族的表达与肝癌的诊断、治疗的关系,进而评价该基因家族作为肝癌免疫治疗靶点和多价肿瘤疫苗研制的可行性。方法对28例肝细胞癌和癌旁组织、3例肝硬化组织以及肝癌细胞株7404、HePG2、7721采用逆转录多聚酶链反应(RT-PCR)方法检测癌-睾丸抗原SSX基因在mRNA水平的表达情况。结果 SSX-1、SSX-2、SSX-3在肝癌癌旁组织和肝硬化组织中均不表达。在收集的肝细胞癌组织中SSX-1、SSX-3基因表达阳性率较高(53.6%、57.1%),SSX-2表达阳性率相对较低(35.7%),其中SSX-1、SSX-2、SSX-3三种基因共同表达的有6例(21.4%),至少有两种SSX基因表达的有23例(82.1%)。结论癌-睾丸抗原某些SSX基因在肝细胞癌中高频率表达与肝癌的发生、发展有关,同时基因的共同表达结果又为多价瘤苗的研究提供了实验方向。Objective To observe the mRNA expression of cancer-testis antigen SSX genes in hepatocellular carcino- ma tissues and cell lines, and to explore the relationship among gene expression and diagnosis and treatment, further- more to evaluate the feasibility of taking advantage of their gene products as immunotherapy targets and tumor vac- cines. Methods mRNA expression was determined by reverse transcription polymerase chain reaction (RT-PCR) in 28 cases of hepatocellular carcinoma tissue, 3 cases of cirrhosis tissue and hepatocellular cancer cell lines 7404, HePG2 and 7721. Results SSX-1 ,SSX-2 and SSX-3 genes were not expressed in caner-adjacent tissue or cirrhosis tissue. SSX-1 and SSX-3 were highly expressed in liver cancer tissues, the expression rates were 53.6% and 57.1%, while SSX-2 positive expression was relatively low, with the expression rate of 35.7%. There were 6 cases (21.4%) co-ex- pressing SSX-1, SSX-2 and SSX-3. And at least two kinds of SSX expressions were found in 23 cases. Conclusion The high expression of certain SSX genes is related to the cause and development of hepatocellular carcinoma, and the co-expression of the genes pave the way for the research of muhivalent vaccine.
