摘 要:目的利用表皮干细胞与结膜去细胞基质膜体外构建复合基质膜组织,移植于实验动物眼表,评价其在结膜重建应用的可行性及价值。方法新西兰白兔20只,随机分为4组,完全剪除右眼球结膜,并在上方巩膜处植入体外构建的表皮干细胞复合基质膜组织,分别于1、2、3、4周后取移植的复合基质膜组织行冰冻切片,标本分别行PAS染色、人结膜上皮细胞特异性标记蛋白K13抗体、MUC5A/C抗体免疫组化鉴定。结果体外移植后的复合基质膜组织在移植后2、3、4周K13抗体、MUC5A/C抗体表达阳性,并在3、4周呈杯细胞PAS染色阳性。结论利用表皮干细胞与结膜去细胞基质膜共建的复合基质膜组织,在临床结膜重建治疗中可作为一种新型的替代移植物。Objective To determine the outcomes of conjunctival reconstruction by transplanting of epidermal progenitor cell - derived artificial conjunctiva in rabbits, further to evaluate the utility of artificial conjunctiva in ocular sur- face reconstruction. Methods A total of 20 New Zealand albino rabbits were divided into 4 groups in random. The bulbar conjunctiva in the right eye was removed and replaced with the artificial conjunctiva graft on the superior superficial sclera area. After 1, 2, 3, and 4 weeks, the artificial compound membranes were collected. Immunohistochemistry with human KI3, MUCSA/C antibody corresponding to the specific surface marker to human conjunctival epithelium, and PAS stain were carried out. Results After 2 weeks of the transplantation, positive human K13 antibody and human MUCSA/C anti- body were observed on compound membranes. Furthermore, positive PAS was observed on goblet cell on the compound membranes. Conclusion The epidermal progenitor cell - derived artificial conjunctiva can be used as a new substitute for clinical conjunctival reconstruction.
