摘  要:采用基因枪法,将SCaM4-Citrine、PepA-mRFP和ECBP21-mRFP3种融合蛋白基因转化到洋葱内表皮细胞.同时,将SCaM4-Citrine、PepA-mRFP基因以及SCaM4-Citrine、ECBP21-mRFP基因分别等量混合,共转化入洋葱内表皮细胞,23℃光照培养18~22h,利用激光共聚焦显微镜可观察到大量融合蛋白的表达,基因枪转化技术获得成功.并发现,对于洋葱表皮细胞来说,幼嫩部分的细胞转化率比老的高,并且枪击压力为7580kPa或9300kPa时为最适压力.Gene gun method is successfully used to transform three syncretic protein genes named SCaM4-Citrine gene, PepA-mRFP gene and ECBP21-mRFP gene into the cuticle of onion. Synchronously, the same proportion taking from the SCaM4-Citrine gene and PepA-mRFP gene as well as the SCaM4-Citrine gene and ECBP21-mRFP gene are transformed into the cuticle of onion, then cultivated for 18-22 hours at 23 ℃. Then large quantities of syncretic proteins can be seen with confocal microscope. The results suggest that the gene gun method can be successfully used. Besides, in our experiments, we find that for onion, young tender cells have higher transformation rate than the older ones, and the most suitable pressure for gun attack is 7 580 kPa or 9 300 kPa.
