摘 要:目的观察中药复方861合剂对肝星状细胞c.fos和c—jun基因表达的影响。方法在培养的肝星状细胞株中分别加入不同浓度的复方861合剂(终质量浓度1.00,0.50,0.25g/L);于8、24、48、72h4个时间点分别收集细胞,提取肝星状细胞总RNA;用逆转录.聚合酶链反应(RT—PCR)方法测定c—fos和c-jun的基因表达水平。结果复方861合剂3组HSC—T细胞c—fos基因表达水平在8h(0.13±0.04、0.20±0.05、0.36-t-O.09)、24h(0.10±0.03、0.16-t-O.04、0.30±0.05)、48h(0.08±0.02、0.12±0.03、0.26±0.04)、72h(0.06±0.01、0.09±0.02、0.21±0.03)4个时间点均明显低于对照组(0.63±0.13、0.50±0.12、0.43±0.06、0.32±0.04);c-jun基因表达水平在8h(0.20±0.04、0.36±0.06、0.53±0.10)、24h(0.17-4-0.04、0.32±0.07、0.47±0.10)、48h(0.13-t-O.03、0.30±0.06、0.40±0.08)、72h(0.10-t-O.02、0.23±0.03、0.32±0.04)4个时l司点也均明显低于对照组(0.93±0.13、0.80±0.12、0.63±0.10、0.32±0.04)。随剂量加大,HSC-T细胞c—fos、c—jun基因表达水平逐渐降低;3组间差异有统计学意义(P〈0.01)。结论复方861合剂对肝星状细胞c—fos和c—jun基因表达有明显的下调作用。Objective To investigate the effects of herbal compound 861 on the expression of c-fos and c-jun genes in hepatic stellate cells. Methods Hepatic stellate cells (HSCs) were cultured in medium containing different concentrations of compound 861 ( 1.00, 0. 50, 0. 25 g/L respectively) and harvested at different incubation periods (8, 24, 48, 72 h). Total RNA of HSCs was isolated and the expression lev- els of c-fos and c-jun genes were detected by using reverse transcription polymerase chain reaction. Results C-fos gene expression levels of HSCs in high (1.00 g/L), middle (0. 50 g/L) and low (0. 25 g/L) con- centrations of compound 861-treated groups were significantly lower than those in control group (0. 63 + 0. 13, 0.50±0. 12, 0.43 +0.06, 0.32 ±0.04) after exposure to compound 861 at 8 h (0. 13 :t:0.04, 0.20±0.05, 0.36 ±0.09), 24 h (0. 10 ±0.03, 0. 16 ±0.04, 0.30 ±0.05), 48 h (0.08 ±0.02, 0. 12 ±0. 03, 0. 26 ±0. 04) and 72 h (0. 06 ±0.01, 0. 09 ±0. 02, 0. 21 ±0. 03). C-jun gene expression levels of HSCs in high, middle and low concentrations of compound 861-treated groups were also signifi- cantly lower than those in control group (0. 93 ±0. 13,0. 80 ±0. 12, 0. 63 ±0. 10, 0. 32 :tO. 04) after ex- posure to compound 861 at 8 h (0. 20 ±0. 04, 0. 36 ±0. 06, 0. 53 ±0. 10), 24 h (0. 17 ±0.04, 0. 32 ± 0. 07, 0. 47 +0. 10), 48 h (0. 13 ±0. 03, 0. 30 ±0. 06, 0.40 ±0.08) and 72 h (0. 10 ±0. 02, 0. 23 ± 0. 03, 0. 32 ±0. 04). The expression levels of c-fos and c-jun genes in HSCs were decreased gradually as the dosage of compound 861 increased, and there were significant differences in the expression levels of c- fos and c-jun genes among three compound 861-treated groups. Conclusion Compound 861 may strongly down-regulate the exlaression of c-los and c-iun ~enes in HSCs.
