摘 要:目的观察骨质疏松对磨损颗粒诱导骨溶解的影响。方法将42只6月龄雌性SD大鼠随机分成A、B和C共3组,A组和B组各18只,C组6只。A组大鼠切除双侧卵巢;B组暂不处理;C组分离双侧卵巢,但不切除。饲养3个月后,从A组和B组中各随机选取6只及C组的全部大鼠,处死后行全身骨密度检查,并取右侧胫骨行显微CT(IxCT)和骨形态计量学检查。A组中剩余的12只大鼠随机分成A1和A2组,每组6只,A1组大鼠颅骨正中矢状缝骨外膜区域,注入5mg的金属钛颗粒悬浮液;在A2组植入等体积的磷酸盐缓冲液(PBS);同法将B组分为B1和B2组。14d后处死所有剩余大鼠,对大鼠颅骨行la,CT及病理切片以检测颅骨溶骨效应。结果与B组和C组比较,A组的骨密度和胫骨骨形态计量学指标显著降低(P〈0.05),胫骨IxCT检查示骨小梁稀疏。A1组和A2组大鼠颅骨行la,CT检查溶骨不明显,病理切片检查示A1、A2、B1和B2组矢状缝骨外膜区域内溶骨面积分别为(0.262-I.-0.009)、(0.130±0.013)、(0.307±0.013)、(0.178±0.011)rain。,组间差异有统计学意义(P〈0.05)。结论在动物模型中,骨质疏松能减缓磨损颗粒诱导的骨溶解过程。Objective To study the influence of osteoporosis (OP) on wear particles-induced os- teolysis in an animal model. Methods Forty-two female Sprague-Dawley (SD) rats aged 6 months were randomly divided into 3 groups: A, B and C. Groups A and B contained 18 rats each, and group C con- tained 6 rats. The rats in group A were operated by bilateral ovariectomy, no operation was done on rats of group B, and that in group C was sham. After 3 months, 6 rats in group A, 6 rats in group B and all the rats in group C were harvested. Bone mineral density ( BMD), txCT and bone histomorphometry were ana- lyzed. The rest of rats in group A were randomly divided into two subgroups : subgroup AI and subgroup A2, and the same as group B. Five mg titanium particles were implanted onto the calvaria of subgroups A1 and B1, and PBS was injected to subgroups A2 and B2. Calvarias were harvested after 14 days and ana- lyzed by jxCT, and histomorphometry was done to measure the osteolysis area of calvarial sagittal suture. Results As compared with groups B and C, BMD and bone histomorphometry index in A group were sig- nificantly reduced (P 〈 0.05 ). The area of calvarial sagittal suture osteolysis was ( 0. 262 ± 0. 009 ), (0. 130 ± 0. 013), (0. 307 ± 0. 013) and (0. 178 ± 0. 011 ) mm2 in subgroups A1, A2, B1 and B2 re- spectively. There was significant difference among the groups. Conclusion Ostoporosis may reduce the particles-induced osteolysis in a rat model.
