摘  要:目的:确定胱天蛋白酶-8(caspase-8)在UVB诱导皮肤成纤维细胞凋亡中的作用。方法:人皮肤成纤维细胞经100、200、300和400 mJ/cm^2 UVB照射后48 h,用MTT法检测细胞活性变化,用Hoechst33258染色法检测细胞凋亡,并用抑制剂Z-IETD-FMK抑制caspase-8活性后,检测凋亡细胞数量。结果:UVB照射明显抑制成纤维细胞活性,引起细胞凋亡,随着照射UVB剂量的增加,凋亡细胞逐渐增多。Caspase-8抑制剂Z-IETD-FMIK抑制UVB照射引起的细胞凋亡。结论:caspase-8在UVB照射导致的UVB急性光损伤中发挥重要作用。Objective: To determine the role of caspase- 8 in the apoptosis of human skin fibroblasts induced by UVB irradiation. Methods: Primary human skin fibroblasts were cultured and were irradiated with 100, 200, 300 and 400 mJ/cm^2 UVB. The viability of the cells was measured by MTT assay at 48 hours after irradiation. The apoptosis of the fihrohlasts was measured with Hoeehst 33258 staining, and was re - measured after the viability of caspase - 8 was inhibited by adding of Z - IEFD - FMK inhibitor. Results: The UVB irradiation markedly inhibited the viability of human skin fibroblasts and induced apoptosis in a dose - dependent manner. After adding of Z - IETD - FMK (the caspase - 8 inhibitor), the apoptosis induced by UVB irradiation was inhibited. Conclusion: Caspase - 8 may play an important role in the process of acute photodamage.
