摘 要:目的:检测独活寄生汤含药血清对成骨细胞OPG/RANKL蛋白表达的影响,从分子生物学层面探究独活寄生汤治疗骨质疏松症的机理,为独活寄生汤治疗骨质疏松症提供理论及实验依据。方法:体外培养原代SD新生24 h大鼠成骨细胞,分别用生理盐水血清对照组、独活寄生汤含药血清低浓度,中浓度,高浓度作用于成骨细胞,采用MTT法检测独活寄生汤含药血清对成骨细胞生长的影响,Western B1ot检测独活寄生汤含药血清对成骨细胞OPG、RANKL蛋白表达的影响。结果:独活寄生汤含药血清以浓度及时间的依赖方式促进成骨细胞生长;Westem B1ot结果显示不同浓度的独活寄生汤含药血清能促进成骨细胞OPG蛋白的表达,并且抑制成骨细胞RANKL蛋白的表达。结论:独活寄生汤含药血清可以通过OPG、RANKL系统促进成骨细胞的增殖并且影响成骨细胞的分化。Objective To investigate the effects of serum containing substances of a Chinese traditional drug,Duhuojisheng Decoction,on expression of osteoprotegerin(OPG) and receptor activator of NF-κB ligand(RANKL) in osteoblasts,and to study the mechanism of action of Duhuojisheng Decoction treatment for osteoporosis.Methods Osteoblasts were obtained from new bom rat calvaria by collagenase-trypsin digestion.Four groups were included in the experiment and situ hybridization method was used to detect the expression of OPG and RANKL.Reaults Compared with the normal serum group,the expression of OPG in osteoblasts in the serum of Duhuojisheng Decoction increased significantly,while the expression of RANKL decreased remarkably.Conclusion The serum containing Duhuojisheng Decoction could not only inhibit the secretion of RANKL to repress the activity of osteoclast but also stimulate osteoblasts to secrete OPG,then the combination of OPG and RANKL increased,which could reduce the activity of RANKL and decresed the activity of the bone resorbing activity of obsteoclasts consequently.It may be one of the mechanism of actions of Duhuojisheng Decoction for treating osteoporosis.
