HIF-1与COX-2在无排卵性功血中的表达及意义

摘　　要：目的检测缺氧诱导因子-1(hypoxia-inducible factor-1,HIF-1)和环氧合酶-2(cyclooxygenase-2,COX-2)在无排卵性功血患者子宫内膜的表达情况,探讨HIF-1和COX-2与无排卵性功血的关系及相关性。方法选取哈尔滨医科大学附属第二医院病理科无排卵性功血中各增生病理类型的存档蜡块70例和正常子宫内膜(因子宫肌瘤行子宫切除的患者)10例。应用SP免疫组织化学方法测定HIF-1及COX-2的表达情况。结果 HIF-1和COX-2在无排卵性功血中阳性染色均定位于子宫内膜腺上皮细胞的细胞质和细胞核中;HIF-1在正常子宫内膜和无排卵性功血中的阳性表达率分别是10.00%(1/10)、42.86%(30/70)。COX-2在正常子宫内膜和无排卵性功血中的阳性表达率分别是20.00%(2/10)、67.14%(47/70)。HIF-1和COX-2在实验组和对照组表达均有显著差异(P<0.05)。HIF-1和COX-2在无排卵性功血中的表达两者之间无相关性(P>0.05)。结论 HIF-1和COX-2均在无排卵性功血中表达增强,二者与无排卵性功血的发生发展相关。HIF-1可能在无排卵性功血子宫内膜血管生成中发挥重要作用,COX-2与抑制子宫内膜腺细胞的凋亡有关及血管生成有关。Objective：To investigate the relationship between HIF-1 and COX-2 by detecting their expression in anovulatory dysfunctional uterine bleeding（ADUB）.Methods：70 cases of various proliferative endometrial samples of ADUB and 10 normal endometrial samples were chosen.All of the cases were obtained from the second affiliated hospital of HMU.The expression of HIF-1 and COX-2 was detected by using immunohistochemical staining with appropriate antibodies.Results：HIF-1 and COX-2 were positively expressed in the nucleus and plasm of glandular epithelial cells of the endometrium;The positive expression rates of HIF-1 in endometrium of normal and ADUB were respectively 10.00%（1/10） and 42.86%（30 /70）,while COX-2 20.00%（2/10） and 67.14%（47/70）.There was significant difference in the expression of HIF-1 and COX-2 between ADUB and normal proliferative endometrium（P0.05）.There was no significant relationship between Livin and Bax.Conclusions：The expression of HIF-1 and COX-2 is potentized in ADUB.They maybe play roles during the development and progression of ADUB.HIF-1 may be have important function during the angiogenesis of ADUB.COX-2 is related to the inhibition of endometrial glandular cell apoptosis and angiogenesis.
