摘 要:目的观察补肾活血中药对大鼠慢性高眼压(elevated intraocular pressure,EIOP)模型视神经损伤的干预作用,探讨其作用机理。方法 30只(30眼)SD大鼠采用烙闭上巩膜静脉法建立大鼠慢性EIOP模型,随机分为三组:对照组、模型组、给药组,每组各10只,观察补肾活血中药对EIOP大鼠眼压(intraocular pressure,IOP)、视神经病理形态学变化以及超微结构的影响。结果本实验采用的烙闭上巩膜静脉法使大鼠IOP明显升高(均为P<0.01),在造模后8周(实验结束)IOP仍为造模前的2~3倍;视神经病理切片半定量分析表明:视神经纤维髓鞘总面积、平均光密度值及积分光密度值分别为:模型组(98048.810±8139.059)μm2、(517.840±118.862)、(18031.320±1895.210),对照组各指标分别为(144975.320±10341.487)μm2、(768.700±167.331)、(29283.350±2331.446),两组间比较差异均有统计学意义(均为P<0.05);给药组各指标分别为(109420.500±55263.250)μm2、(710.080±150.956)、(24225.730±5455.363),与模型组各指标比较,差异均有统计学意义(均为P<0.05);模型组大鼠视神经超微结构损害明显,给药组损害有所改善。结论补肾活血中药有助于EIOP大鼠视神经损伤的修复。Objective To observe the effects of traditional Chinese medicine ( TCM) of Bushenhuoxue on the pathological changes of optic nerve injury in rat model of chronic elevated intraocular pressure ( EIOP) ,and preliminarily explore its mechanism.Methods By unilaterally cauterizing 30 episcleral vessels,the rat model of chronic EIOP was established.It was randomly and equally divided into 3 groups: control group,model group and treatment group,10 cases in each group.After giving TCM of Bushenhuoxue and normal saline for 8 weeks,the rats were killed and taken out the brains,and the effects of Bushenhuoxue on intraocular pressure ( IOP) and pathological changes of optic nerve were observed.Results IOP of rat model obviously increased by unilaterally cauterizing episcleral vessels( all P 0.01 ) .At the end of the experiment,IOP was 2 - 3 times than before modeling; Semi-quantitative pathological analysis showed that the total area of myelin sheathin in optic nerve fiber,average optical density and integrated optical density in model group were ( 98 048.810±8139.059) μm 2 ,517.840±118.862,18 031.320±1895.210 were significant different from the control group [( 144 975.320±10 341.487 ) μm 2 ,768.700±167.331, 29 283.350±2331.446,all P 0.05 ]; There were statistical differences between model group and treatment group [( 109 420.500±55 263.250 ) μm 2 ,710.080±150.956,24 225.730±5455.363,all P 0.05 ]; There were obvious ultrastructural damages in optic nerve of model group,which reduced in treatment group.ConclusionTCM of Bushenhuoxue can repair the injury of optic nerve in the rat model of chronic EIOP.
