摘 要:目的:优选大孔吸附树脂分离、纯化葛根总黄酮和葛根素的工艺。方法:考察AB-8,D-101,HPD-100,S-8 4种不同极性吸附树脂对葛根中总黄酮和葛根素的静态吸附及解吸性能,筛选树脂型号;以葛根素、总黄酮转移率和纯度为指标,通过单因素试验考察其吸附和洗脱条件。结果:选用AB-8型大孔吸附树脂,优选的工艺条件为上样液质量浓度1.0 g.mL-1,葛根与大孔树脂质量比1∶2,径高比1∶6,用2 BV水洗除杂,加2 BV 80%乙醇以2 mL.min-1洗脱。转移率均>92%,所得葛根总黄酮纯度达80%,葛根素质量分数25%。结论:AB-8型大孔树脂用于富集葛根总黄酮和葛根素效果最佳,是一种理想的分离纯化介质,且优选的工艺操作简单、方法可靠。Objective: To optimize separation and purification technology of total flavonoids and puerarin from Pueraria lobata by macroporous adsorption resin. Method: Static adsorption and desorption performance of four kinds of different polar macroporous adsorption resins (S-8, AB-8, D-101, HPD-100) for total flavonoids and puerarin from P. lobata were investigated, in order to select optimal resin; With yield and purity of total flavonoids and puerarin as indexes, single factor test was used to investigate adsorption and elution conditions. Result: AB-8 macroporous resin was chosen, its optimized technology conditions were as followings: the concentration of sample solution 1.0 g mL-1, mass ratio of raw material-resin 1 : 2, diameter-height ratio of column 1: 6, washed by 2 BV water, eluted with 2 BV 80% ethanol, elution flow rate 2 mL min-1 Elution rate of total flavonoids and puerarin were more than 92% , purity of total fiavonoids and puerarin could reach 80% and 25%, respectively. Conclusion: AB-8 macroporous adsorption resin was sutiable for enrichment total flavonoids and puerarin from P. lobata, it was a kind of ideal separation and purification medium, optimized technology was simple and reliablel
