摘 要:目的:优选黄连提取液的超滤纯化工艺参数。方法:以出膏率及盐酸小檗碱、巴马汀、黄连碱及表小檗碱含量为指标,通过单因素试验考察截留相对分子质量分别为5 000,10 000,20 000的中空纤维膜对黄连提取液超滤工艺的影响;选取料液温度、料液质量浓度及进样流速为考察因素,以指标成分的综合含量和转移率的综合评分为指标,采用正交试验优选黄连提取液的超滤工艺参数。结果:截留相对分子质量为5 000的中空纤维超滤膜分离效果较好,其最佳超滤工艺参数为料液温度40℃,体积流量40 mL.min-1,药液质量浓度25 g.L-1。结论:中空纤维超滤膜可选择性地用于药液的精制,可提高有效成分的保留率。Objective: To optimize ultrafiltration purification technology parameters of Coptis chinensis extract. Method: With extract rate, the content of herberine hydrochloride, coptisine, palmatine and epiberberine as indexes, influence of hollow-fiber membranes with relative molecular weight 5 000, 10 000 and 20 000 on ultrafiltration purification technology of C. chinensis extract was investigated by single factor test; Temperature and concentration of sample liquid, sample flow rate were chosen as factors, orthogonal test was used to optimize uhrafiltration purification technology parameters of C. chinensis extract with composite score of integrated content and transfer rate of index components as an indicator. Result: Hollow-fiber membrane with 5 000 relative molecular weight had better separation efficiency as others, optimum ultrafiltration technology parameters were as followings : liquid temperature 40 ℃ , liquid speed 40 mL min - 1 , liquid concentration 25 g. L- 1o Conclusion : Hollow-fiber membrane could be selectively used in purification of liquid, it could increase retention rate of active ingredient.
