摘  要:目的分析比较磁珠法和滤膜法提取病毒核酸的重复性及灵敏度。方法选用国产的磁珠法和滤膜法试剂同时提取核酸检测临床手足口病标本,荧光相对定量比较两种方法的重复性及灵敏度。结果磁珠法提取核酸的标本用量较少,成功率较高,磁珠法试剂检测重复性较好,灵敏度较高。结论磁珠法提取核酸在手足口病的荧光定量检测中优于滤膜法。Objective To investigate the repeatability and sensitivity of the magnetic bead and filter extracting reagents for extracting nuclear acids. Methods The magnetic bead and filter extracting reagents(domestic) were used in extracting the nuclear acids from HFMD clinical specimens.Compared repeatability and sensitivity of the two reagents by real-time relatively quantify RT-PCR. Results For extracting the nuclear acids,the magnetic bead reagent had consumed less specimens,and had higher success rate.There were better repeatability(CVmagnetic bead<CVfilter membrane)and sensitivity(ΔΔCt magnetic bead<ΔΔCt filter membrane)of the magnetic bead reagent. Conclusion The ways that extract nuclear acids from HFMD clinical specimens with magnetic bead reagent(domestic) was better than filter membrane reagent(domestic).
