摘  要:目的了解2010年贵州省H3N2亚型流感病毒HA1基因变异特征。方法用MD-CK细胞分离流感病毒,采用血凝抑制试验分型鉴定,随机选取8株H3N2亚型流感毒株进行RNA提取,逆转录扩增获得HA1基因产物,将其进行基因产物测序,测定的序列用相关的生物信息软件进行分析。结果贵州省各代表株在同一分支。该分支与2008-2009年疫苗推荐株A/Brisbane/10/2007(H3N2)接近,与WHO 2010年疫苗推荐株A/Perth/16/2009(H3N2)相距较远。结论2010年根据WHO疫苗推荐株生产的疫苗对该年我市H3N2亚型流感预防效果不好,以至于H3亚型在我省成为优势株,引起广泛流行。Objective To understand genetic variation characteristics of HA1 gene of influenza vi- rus H3N2 subtype isolated in Guizhou in 2010. Methods Influenza viruses isolated were cultured in MDCK cell and subtype was tested by hemagglutination inhibition. There were 8 strains chosen ran- domly to be tested. Viral RNA was extracted and their HA1 genes were amplified by reverse tran- scription polymerase chain reaction (RT-PCR). The PCR products were sequenced, and the gene se- quences were analyzed through biometic software. Results The 8 strains which isolated form Guizhou in 2010 belong to the same lineage. The sequences of 8 strains were similar to A/Brisbane/10/2007 (2008--2009) vaccine strains and far from A/Perth/16/2009(2010) vaccine strain recommended by WHO, it was not in the same branch of a tree. Conclusion The H3N2 subtype vaccine strain recom- mended in 2010 by WHO had no effective protection, which was the main reason for why the H3N2 virus was dominant in 2010.
