摘 要:目的分析非嗜酸细胞性哮喘患者Th17相关细胞因子白细胞介素(IL)-8、IL-17和IL-23受体(IL-23R)水平,探讨其在非嗜酸细胞性哮喘发病机制中的作用。方法以24例非嗜酸细胞性哮喘患者和25例健康者(对照组)为研究对象,3.0%高渗生理盐水10 ml超声雾化吸入行痰诱导,抽空腹肘静脉血5 ml,离心后取痰上清液及血清,采用酶联免疫吸附试验(ELISA)法测定诱导痰上清液及血清中IL-8、IL-17和IL-23R的浓度。Wright’s染色观察痰液细胞涂片中炎症细胞的浸润情况。结果与对照组相比,非嗜酸细胞性哮喘患者痰液细胞涂片中示大量炎症细胞浸润,以中性粒细胞和淋巴细胞为主,其血清和诱导痰上清液IL-8、IL-17和IL-23R水平均明显增高,差异均有统计学意义(P<0.05,P<0.01)。结论 Th17相关细胞因子IL-8、IL-17和IL-23R在非嗜酸细胞性哮喘发病机制中具有重要作用。Objective To evaluate the expression levels of interleukin-8 (IL)-8, IL-17 and IL-23 receptor(IL-23R) in patients with non-eosinophilis asthma (NEA) and to investigate the roles of above interleukins in the NEA development. Methods The serum and sputum supernatant samples were from 24 case with NEA and 25 healthy controls. The expression levels of IL-8, IL-17 and IL-23R in serum and sputum supernatant samples were detected with enzyme-linked immunosorbent assay (ELISA). The airway inflammation was observed with sputum smear by Wright's staining. Results Compared with healthy controls, more inflammatory ceils in sputum smear of patients were observed and the expression levels of IL-8, IL-17 and IL-23R in the serum and sputum supernatant of patients significantly increased(P〈0.05 or P〈0.01 ). Conclusion Cytokines(IL-8,IL-17 and IL-23R) related to Thl7 may play important roles in the development of non - eosinophilis asthma.
