摘  要:目的:测定新疆赤芍多糖中半乳糖醛酸的含量。方法:采用间羟基联苯法对新疆赤芍多糖中的半乳糖醛酸进行含量测定,并研究四硼酸钠-浓硫酸溶液用量、加酸后沸水浴时间、间羟基联苯溶液用量、显色时间及中性糖对测定结果的影响。结果:最佳测定条件为:四硼酸钠-浓硫酸溶液用量5.0mL,沸水浴加热5min,间羟基联苯用量为80μL,显色时间10min,测定波长为525nm。新疆赤芍多糖中糖醛酸的平均含量为24.49%。结论:间羟基联苯法适用于新疆赤芍多糖中半乳糖醛酸的含量测定。Objective:To determine the galacturonic acid content of the Polysaccharides from Radix Paeoniae hybridae. Methods: The method with Meta-hydroxydiphenyl was used. The amount of Na2B4OT-H2SO4, the heating time, m-hydroxydiphenyl, the color development time and the interference by neutral sugar were studied. Results: The optimized parameters were obtained as fol-lows: the dosage of Na2B407-H2SO4 and m-hydroxydiphenyl was 5.0 mL and 80μL, time for heating and color development were 5 min and 10 min respectively. The detection wavelength was 525nm. The average content of galaeturonie acid in Polysaceharides from Radix Paeoniae hybridae is 24.49 %. Conclusion:The method of m-hydroxydiphenyl can be well used for the determination of galacturonic acid in Polysaecharides from Radix Paeoniae hybridae.
