摘  要:针对单株DNA、多株叶片混合DNA和单株DNA混合的DNA取样方法,利用SSR技术对蒙A群和C群1进行遗传变异分析,探讨建立玉米群体遗传多样性分析的技术体系。采用均匀分布在玉米10条染色体上的34对SSR引物对不同处理的DNA样本进行扩增,通过聚丙烯酰胺凝胶电泳检测,对60个单株进行多态性信息量与遗传相似系数分析,比较不同处理的DNA样本扩增的等位基因数目。结果表明,2个供试群体均具有较丰富的遗传变异,建立的技术体系可用于群体的遗传多样性研究,采用12株叶片混合、5次重复提取的DNA样本可以代替相同数目的单株DNA混合的DNA样本,为最优取样方法。DNA mixing sampling methods, which extracted from muhi-plant-leaf DNA, single-plant-leaf DNA and single-plant DNA were used to analyze the genetic variations of the Meng A and Meng C populations using SSR markers and discuss the optimum technique system for analyzing genetic diversity of maize. DNA samples from these treatments were propagated using 34 SSR primers which homogeneous distributed in the 10 chromosomes of maize. Amount of information about polymorphism and the index of genetic diversity of these 60 plants were analyzed, and the numbers of alleles of the propagating DNA samples were also compared using polyacrylamide gel electrophoresis. The results indicated that these two maize populations have relative rich genetic variations and the new built technique system was able to do the research on maize genetic diversity. The method, which the DNA samples extracted from 12 plants mixing leaves with five repeats, is the best sampling method. This sampling method is able to be used to analyze the genetic relationships among the plentiful maize populations, and significantly increase work efficiency.
