多指标综合评价优选蟾皮脂溶性成分提取工艺

摘　　要：目的考察蟾皮中的脂溶性有效成分华蟾酥毒基和酯蟾毒配基的乙醇回流提取工艺。方法建立HPLC法测定蟾皮提取物中两种有效成分的含量,采用正交试验设计,以乙醇浓度、提取时间及加醇倍数为因素,以浸膏得率、两种有效成分含量为评价指标,考察蟾皮乙醇煎煮回流提取方法,结合方差分析确定最佳提取工艺。结果在0.35～5.6μg·ml-1范围内,华蟾酥毒基线性方程:A=36 793 C+743.9(r=0.9990,n=6),平均加样回收率为97.77%;在0.30～4.8μg·ml-1范围内,酯蟾毒配基回归方程:A=28 149 C+2084.5(r=0.9990,n=6),平均加样回收率98.31%。乙醇煎煮回流法各因素影响顺序为:乙醇浓度﹥提取时间﹥加醇倍数,确定最优提取工艺为A3B1C1。结论乙醇浓度是影响蟾皮中脂溶性成分提取工艺的最主要因素,采用多指标综合评价法优化蟾皮脂溶性成分提取工艺合理、可行。Objective To study an ethanol extraction technology of cinobufagin and resibufogenin from the skin of Bufo bufo gargarizans. Methods HPLC method was established to determine the content of cinobufagin and resibufogenin from the skin of Bufo bufo gargarizans. Orthogonal experiment design and variance analysis were employed to optimize extraction conditions. The independent variables selected were ethanol concentration, extraction time and the amount of alcohol added while the dependent variables selected were the rate of extraction, content of einobufagin and resibufogenin. Results The linear range of cinobufagin and resibufogenin was within 0. 35-5.6 μg ·ml^-1 and 0. 30-4. 8 μg ·ml^-1. The regression equation wasA =36793 C ＋743.9（ r =0.9990, n =6）andA =28 149 C ＋2084.5（ r =0. 9990, n =6）. The average recovery was 97.77% and 98.31%. The most important factor of influence was ethanol concentration,followed by the extraction time and alcohol added. Optimum technology was A3 B1C1. Conclusion Ethanol concentration is most important for extraction of liposoluble constituents from the skin of Bufo bufo gargarizans cantor. Optimizing extraction conditions using multi-index comprehensive evaluation method is feasible.
