摘  要:目的:通过观察苏木乙酸乙酯提取物对健康大鼠外周血细胞因子IL-1、IL-2、IL-12及IFN-γ含量的影响,探讨其免疫抑制的作用机制。方法:健康Wistar大鼠32只,随机分为空白组、高剂量组、中剂量组、低剂量组,每组8只大鼠。高、中、低剂量组分别按照2.3g/kg/d、1.15g/kg/d、0.58g/kg/d剂量给予苏木乙酸乙酯提取物灌胃,空白组给予同体积橄榄油灌胃,连续灌胃7天,眼球取血,采用ELISA法检测大鼠血清IL-1、IL-2、IL-12及IFN-γ含量。结果:苏木乙酸乙酯提取物高、中、低剂量组与空白组比较,IL-1的表达无明显差异;高、中剂量组IL-2、IFN-γ的表达均降低,与空白组比较有显著性差异(P<0.05)。高、中、低剂量组IL-12的表达均降低,与空白组比较有显著性差异(P<0.05)。结论:苏木乙酸乙酯提取物具有免疫抑制作用,其作用机理可能与抑制细胞因子IL-2、IL-12、IFN-γ的表达有关。Objective:To observe the effect of ethyl acetate extract of sappanwood on the level of IL-1,IL-2,IL-12 and IFN-γ in peripheral blood of healthy rats in order to discuss the immunosuppressive function of ethyl acetate extract of sappanwood.Method:32 healthy Wistar rats were randomly divided into four groups which were blank group,high dose group,middle dose group and low dose group.Every group had eight rats.Give high dose group,middle dose group and low dose group rats ethyl acetate extract of sappanwood 2.3g/kg/d,1.15g/kg/d,0.58g/kg/d and give blank group the same volume olive oil.After giving rats four groups ethyl acetate extract of sappanwood or olive oil ceaselessly,we got the blood from rats' eyeballs.Detect content of IL-1,IL-2,IL-12 and IFN-γ in serum of the rats through ELISA method.Result: There was no obvious difference between high dose,middle dose,low dose and blank dose groups in the influence of ethyl acetate extract of sappanwood on the level of IL-1 in peripheral blood of healthy rats;High dose and middle dose groups had an obvious difference between blank group in expression of IL-2 and IFN-γ,which decreased the expression in serum of high dose and middle dose groups(P0.05).High dose,middle dose and low dose groups had obvious difference between blank group in expression of IL-12 decreased the expression in serum of high dose,middle dose and low dose groups(P0.05).Conclusion: Ethyl acetate extract of sappanwood has an immunosuppressive function.The mechanism maybe has a relationship with restraining the expression of IL-1,IL-2,IL-12 and IFN-γ.
