摘 要:目的研究热CO2气腹对COLO205细胞株侵袭迁移能力及转移相关蛋白-基质金属蛋白酶-2表达的影响。方法 COLO205细胞分别经热CO2气腹43℃作用3~4h,体外迁移和侵袭实验评价其对细胞侵袭迁移能力的影响;并利用同位素标记的相对和绝对定量技术标记,液相色谱分离蛋白质,质谱仪找出处理前后表达差异蛋白,同时利用蛋白质印迹和RT-PCR进行验证。结果热气腹不仅对COLO205细胞侵袭和迁移具有抑制作用,并呈作用时间依赖性;而且对COLO205细胞株MMP-2的表达有抑制作用,与作用时间相关。结论热CO2气腹能抑制COLO205的侵袭和迁移,其机制可能与热CO2气腹抑制了细胞内的MMP-2的表达有关。Objective To investigate the influence of hyperthermic CO2 pneumoperitoneum on the biological behaviors and the expression of metalloproteinase-2 of human colorectal cancer cells. Methods COLO 205 cells were treated by hyperthermic CO2 pneumoperitoneum (HT-CO2) at 43℃ for 3 to 4 hours. The effects of HT-CO2 on the biological behaviors of COLO 205 cells were also measured by invasion and metastasis assay in vitro. Differential proteins were identified by quantitation assay using isotope labeling and confirmed by Western blot and RT-PCR. Results HT-CO2 treatment inhibited invasion and metastasis of COLO 205 cells in a time-dependent way, and at the same time decreased the expression of metalloproteinase-2. Conclusion HT-CO2 could inhibit the invasion and metastasis ability of COLO 205 cells, and the possible mechanism might be associated with decreased expression of metalloproteinase-2.
