摘 要:目的:在分子水平上对浙产野生白及进行遗传多样性研究,以确定浙产白及生物多样性现状,为有效保护浙产白及种质资源提供理论基础。方法:采用简单重复序列区间(ISSR)标记技术对浙江九个不同产地白及进行研究,通过UPGMA聚类进行分析。结果:从45个ISSR随机引物中筛选出12条引物,共扩增出164条条带,其中多态性条带143条,多态性达87.19%,品种之间的遗传相似系数范围在0.59~0.76间;聚类分析结果显示九个浙产白及分为两大分支:杭州、宁波和德清为一支,淳安、天台、温州、临安、嘉兴、衢州为另一支。结论:浙产白及遗传多样性较低,可引进外来品种或通过杂交育种的方式丰富浙产白及种质资源,并需要对白及种质资源进行有效的保护和合理的利用。Objective:In order to ensure the diversity situation of wild Bletilla striata grown in Zhejiang province by the genetic experiment at molecular level and supply theoretical foundation for protecting its species diversity. Method:Nine samples collected from different places of Zhejiang province were analyzed by Inter - Simple Sequence Repeat(ISSR) mo- lecular marker technology , and cluster analysis was made through UPGMA method. Result:Twelve primers were screened out from fortyfive primers. Altogether, 164 DNA fragments were got with these 12 primers, 143 fragments were demonstra- ted specific band within these fragments and the polymorphic rate was 87.19% ,the genetic similarity coefficient between species ranged from 0.59 - 0.76, the result of cluster analysis showed that 9 samples were divided into two groups : the samples from Hangzhou, Ningbo and Deqing were classified as one group and Chunhn, Tiantai, Wenzhou, Linhn, Jiaxing, Quzhou as another group. Conclusion:The genetic diversity of the Bletilla grown in Zhejiang province was low, and the germplasm resource of which could he enriched through introducing foreign varieties and crossbreeding,and moreover, ef- fective measurements should he taken to protect and reasonably use the germplasm resources of wild Bletilla striata.
