摘 要:目的:观察补肾活血方补肾活血方对前列腺增生大鼠膀胱组织胶原沉积的影响。方法:清洁级雄性Wistar大鼠48只,随机分为正常组、模型组、模型加补肾活血方组、模型加坦索罗辛组4组。除正常组外,其它实验大鼠以去势加睾酮注射法复制大鼠BPH模型。于去势第一天起,各组大鼠分别给予相应药物灌胃。模型+补肾活血方按1 mL/100(g.d)体重予补肾活血方(由淫羊藿、枸杞子、穿山甲、丹参、菟丝子等组成,含生药量2.34g/mL)灌胃,模型加坦索罗辛组按1.8μg/100(g.d)体重予坦索罗辛灌胃。其余各组予等体积生理盐水灌胃。均每日1次,连续38 d后检测各组大鼠前列腺、膀胱湿重及脏器指数,免疫组织化学法观察膀胱组织内胶原沉积。结果:模型组膀胱胶原Ⅲ阳性面积率、总IOD等均明显高于空白组(P<0.01)。补肾活血方、坦索罗辛组膀胱胶原Ⅲ阳性面积率及总IOD均明显较模型组低(P<0.01)。膀胱胶原Ⅰ沉积情况与胶原Ⅲ类似。模型组膀胱胶原Ⅰ/胶原Ⅲ总IOD可降至0.28,与空白组相比有非常显著的差异(P<0.01),补肾活血方组则明显高于模型组(P<0.01),与空白组相比无统计学差异;结论:补肾活血方组可以明显降低BPH模型大鼠膀胱组织胶原Ⅲ沉积,提高胶原I/胶原III比率,这可能是补肾活血方有效改善BPH继发膀胱改变的机制之一。Objective :To observe the effect of BushenHuoxue Fang of deposition of collagen in rat bladder tissue.
