摘 要:目的:观察肾茶对阿霉素肾病大鼠外周血中核因子-KB(Nuclear factor-KB,NF-KB)、白细胞介素-8(Interleukin-8,IL-8)表达的影响,探讨肾茶治疗肾病综合征(Nephrotic Syndrome,NS)的可能机制。方法:40只Wistar雄性大鼠随机分为两组:正常组(n=10)注射生理盐水0.5mL,肾病组(n=30)尾静脉单次注射阿霉素5mg/kg(用生理盐水溶解至0.5mL)制备NS模型。注射后第7天检测所有大鼠24h蛋白尿,尿蛋白>30mg/24h者为NS模型成功。将模型成功的肾病组再随机分为两组:肾病组(n=10)、肾茶组(n=10)。肾茶组在造模成功后第7天开始灌胃给药5mL/kg,正常组和肾病组给予等量生理盐水灌胃。第7、21、42天测各组大鼠24h尿蛋白定量(免疫比浊法)及外周血单个核细胞提取物NF-KB(酶联免疫吸附法)及其上清液IL-8(酶联免疫吸附双抗体夹心法)的变化。结果:①肾病组24h尿蛋白定量、NF-KB及IL-8含量均高于正常组和肾茶组,有显著性差异(P<0.01)。②肾茶组24h尿蛋白定量、NF-KB及IL-8含量均比肾病组明显下降,有显著性差异(P<0.01)。③肾病组和肾茶组的24h尿蛋白、NF-KB、IL-8两两之间呈直线相关性(P<0.01)。结论:肾茶可能直接或间接抑制NF-KB,降低IL-8水平,减轻尿蛋白,从而延缓疾病进展。Objective:To observe the effects of Clerodendranthus spicatus on nuclear factor--KB (NF-- KB) and interleukin- 8 (interleukin--8, IL--8) expression in peripheral blood of rats with adriamycin nephropathy, and to explore the possible mechanisms of the Clerodendranthus spicatus treatment of nephrotic syndrome (nephrotic syndrome, NS). Methods.Forty Wistar rats were randomly divided into 2 groups, normal group(n= 10) were injected with saline(0. 5mL), Nephropathy group(n=30) received single tail vein injection of adriamycin (with physiological saline solution to 0. 5mL) in 5mg/kg to build NS model. 24hours urine pro-tein was detected in all rats on the Seventh days after injection. Those with urine protein〉30mg/24h became successful NS model. The successful modeling rats were =10), and Clerodendranthus spicatus group (n= 10). randomly divided into 2 groups: Nephropathy group(n The Clerodendranthus spicatus group was given intra- gastric administration in 5ml/kg on 7d after modeling success, and the normal group and nephropathy group were given the same amount of saline lavage. On 7d, 21d, 42d, 24 hours urine protein quantity(Immune tur- bidimetry) , peripheral blood monouclear protein extract NF--KB(ELISA) and supernatant of IL--8( ELISA double antibody sandwich method) of all the rats were tested to find the changes. Results . ①Compared with the normal group and the spicatus group, the quantitate of 24 hour urine protein, NF--KB and IL--8 in the nephropathy group increased significantly (P 〈0.01). ②Compared with the nephropathy group,the quantity of 24 hour urine protein, NF--KB and IL--8 in the spicatus group reduced significantly (P〈0. 01).③The 24 --hour urine protein was linear correlated with NF--KB and IL--8 in nephropathy group and spicatus group. Conclusions:Clerodendranthus spicatus may directly or indirectly inhibit NF--KB, reduce urinary protein, and therefore delay the progression of disease.
