江西部分地区猪痘病毒PCR检测及P35基因的分析

摘  要:为了解江西部分地区猪痘病毒(SWPV)的感染及流行情况,从江西省7个地区(市)共36个规模化猪场采集55份疑似猪痘病猪的皮肤痘痂组织,应用PCR方法对其进行猪痘病毒核酸检测,并随机挑选19个PCR产物进行序列测定和P35基因序列分析。结果表明,55份样品中,有44份样品检测为SWPV阳性,阳性率高达80%;所测的19条序列与SWPV参考毒株(登录号:AF410153.1)P35基因核苷酸同源性为96.3%~98.4%,推导的氨基酸同源性为87.5%~88.5%;各序列之间的核苷酸同源性为97.7%~100%,推导的氨基酸序列同源性为98.2%~100%。结果表明江西省部分地区的猪群中存在猪痘病毒感染。To investigate the prevalence of swinepox virus (SWPV) in Jiangxi province, 55 skin vesicles were collected from 36 pig farms in Jiangxi province for detection of swinepox virus P35'gene by PCR. The results showed that 44 samples were SWPV positive, the positive rate was 80 percent. The homology and phylogenetie tree were analyzed by DNAStar, results indicated that the homologies of nucleotide and deduced amino acid sequences of SWPV P35 gene of these isolates to the SWPV reference strain (AF410153.1)were 96.3%-98.4% and 87.5%-88.5%, and the homologies of nucleotide and deduced amino acid sequences between the isolates were 97.7%-100% and 98.2M-100% respectively.
