摘  要:目的研究白藜芦醇对呼吸道合胞病毒(RSV)感染Hep-2细胞相关周期的影响,并探讨金欣口服液(麻黄、苦杏仁、生石膏、葶苈子、桑白皮、前胡、黄芩、虎杖)治疗RSV肺炎的物质基础。方法构建RSV感染Hep-2细胞体外模型,分为白藜芦醇处理组与未处理组,并以没有感染RSV的正常细胞作为对照组;采用体外转录法、荧光定量PCR法(qPCR)及激光扫描共聚焦显微镜法分别检测白藜芦醇对RSV的RNA聚合酶转录活性、核酸复制及诱导Hep-2细胞融合效应的影响。结果白藜芦醇在25μmol/L的最大无毒浓度下,能显著抑制RSV的RNA聚合酶活性,但对RSV复制、融合蛋白F表达无明显作用,亦不能抑制RSV介导的细胞融合效应。结论白藜芦醇在金欣口服液的抗RSV效应中发挥作用不是通过干预RSV复制相关周期,需要进一步研究。AIM To study the effects of resveratrol on replicative cycle of respiratory syncytial virus (RSV) in the Hep-2 cells, and to investigate the pharmaeodynamic action of Jinxin Oral Liquid (Ephedrae Herba, Armeniacae Semen amarum, Gypsum Fibrosum, Lepiclii Semen, Mori Cortex, Peucedani Radix, Scutellariae Radix, Polygoni cuspidate Rhizoma et Radix). METHODS Hep-2 cell models infected by RSV were divided into two groups. One was treated with resveratrol and another was not treated with resveratrol. Normal cells uninfected by RSV were regarded as control group. The methods, such as transcription in vitro, real time quantitive PCR and immunofluorescence with laser scanning confocal microscopy, were used respectively to detect the RNA polymerase activity, nucleic acid duplication and cell fusion induced by RSV. RESULTS There was no significant effect of resveratrol at the maximum nontoxic concentration of 25 μmol/L on duplication and cell fusion except transcription of RSV. CONCLUSION intervention of RSV Resveratrol plays a role in the anti-RSV effect of Jinxin Oral Liquid, probably not through the replicative related cycle. The mechanism needs further researches.
