摘  要:目的探讨降压益肾颗粒(鬼针草、何首乌、山茱萸、玄参、泽泻、川牛膝等)对转化生长因子(TGF-β1)及相关指标的影响。方法将自发性高血压大鼠分为模型组、贝那普利组、降压益肾颗粒大、中、小剂量组,及正常Wistar大鼠组,每组8只,治疗后第8周和12周分别采集血液,ELISA法测定血清中TGF-β1、内皮素-1(ET-1)水平;实时荧光PCR法测定肾组织中TGF-β1mRNA、Ⅳ胶原(COLⅣ)mRNA表达水平;免疫组化法检测肾脏中TGF-β1、COLⅣ蛋白表达。结果各治疗组血液中TGF-β1、ET-1与模型组比较,第8周和第12周TGF-β1均下降,有显著差异(P<0.05~0.01);各组大鼠肾组织中TGF-β1mRNA、COLⅣmRNA表达水平与模型组比较具有显著差异(P<0.05~0.01);免疫组化结果表明,TGF-β1,COLⅣ表达除模型组明显增强外,各给药组均明显减弱,顺序依次为降压益肾低剂量组、贝那普利组、降压益肾中剂量组、降压益肾高剂量组、正常组,降压益肾高剂量组与正常组相当。结论降压益肾颗粒可能通过减少TGF-β1的分泌、抑制血管收缩因子ET-1,进而抑制肾小球细胞外基质的沉积,改善肾脏硬化,从而达到肾脏保护目的。AIM To explore the effect of Jiangya Yishen Granules (Bidentis pilosae Herba, Polygoni multifiori Radix, Corni Fructus, Scrophulariae Radix, Alismatis Rhizoma, Cyathulae Radix, etc. ) on the expression of transforming growth factor (TGF-β1) and other relative factors. METHODS Spontaneously hypertensive rats (SHR) were randomly divided into four groups, positive model group, low-dose treatment group, middle-dose treatment group and large-dose treatment group. Wistar rats were set as normal group. Each group consisted of eight rats. Their blood was collected at the 8th and 12th week during the treatment. The levels of serum TGF-β1 and endothelin-I (ET-1) were detected by ELISA. The expressions of TGF-β1 mRNA and collagenlV (COL1V) mRNA were detected by PCR. The expressions of TGF-β1, COL1V protein in renal tissue were detected by immunohisto-chemical assay. RESULTS Compared with the model group, TGF-β1 level of all treated groups at the 8th and 12th week decreased significantly (P 〈0. 05 -0. 01 ). There were significant differences in their expressions of TGF-β1 mRNA and COL1VmRNA in the renal tissues (P 〈 0. 05 -0. 01 ). Immunohistochemical result showed that the expressions of TGF-β1, COLIV protein significantly increased in positive model group while decreased in all treated groups. The sequence of protein level in descending order was low-dose treatment group, positive model group, middle-dose treatment group, large-dose treatment group and normal group. Large-dose treatment group proved tobe similar with normal group. CONCLUSION Jiangya Yishen Granules may decease the secretion of TGF-β1 and ET-1, and then inhibit the expanding of extraeelluar matrix, alleviate glomerular sclerosis and thereby protect kidney function.
