摘  要:目的建立大鼠血浆中莪术醇和吉马酮的HPLC测定方法,研究莪术提取物中莪术醇和吉马酮在正常大鼠血浆中的药物代谢动力学特征。方法 SD大鼠经灌胃莪术提取物后,采取不同时间点血样,血浆样品经乙腈沉淀处理后供HPLC分析,其色谱参数为Hypersil ODS柱(250 mm×4.6 mm,5μm),流动相为乙腈(0~20 min:63%~66%)-水,体积流量1 mL/min,柱温25℃,检测波长210 nm。结果莪术醇和吉马酮的线性范围分别为0.025~5μg/mL和0.025~2.5μg/mL,最低检测限均为0.025μg/mL,高、中、低3个质量浓度的质控样本的方法回收率在95.2%~105.3%间,日内、日间精密度(RSD)均小于5%。莪术醇和吉马酮的消除半衰期分别为5.745 h和22.002 h;AUC(0-t)分别为8.814μg.h/L和0.739μg.h/L,两者的血药浓度—时间曲线符合二室模型。结论建立的血浆中同时测定莪术提取物中莪术醇和吉马酮的RP-HPLC测定方法能用于莪术在大鼠体内的药代动力学研究。AIM To develop an HPLC method for simultaneously determining curcumenol and germacrone in rat plasma and to investigate their pharmacokinetics characteristic of zedoary turmeric aqueous extract. METHODS After intragastric administration, SD rats' plasma concentrations of curcumenol and germacrone were collected and deproteinized with acetonitrile. The analysis was then performed on a Hypersil ODS column (250 mm x 4. 6 mm, 5 μm). The mobile phase was composed of acetonitrile (0 -20 rain: 63% -66% ) and water with a linear gradient elution. The flow rate was 1.0 mlMmin and the temperature of column was set at 25 ℃. The UV detection wavelength was set at 210 nm. RESULTS Linear calibration curves fell within the ranges of 0. 025 -5 μg/mL and 0. 025 -2.5 μg/mL, respectively. The lowest detection limit was 0. 025 μg/mL. The average recoveries were 95.2% - 105.3% and the inter- and intra-day RSDs were all less than 5%. Elimination half lives of curcumenol and germacrone were 5. 745 h and 22. 002 h, respectively. The area under the concentration-time curve (AUC0-t ) values were 8. 814 and 0. 739 μg· h/L, respectively. The plasma concentration-time data fit to a two-compartment model, coNcLuSION RP-HPLC method established simultaneously determining curcumenol and germacrone in rat plasma can be applied to the pharmacokinetics studies of Curcumae Rhizoma.
