摘 要:目的研究假体磨损颗粒是否通过核转录因子-κB(NF-κB)信号途径上调巨噬细胞移动抑制因子(MIF)、肿瘤坏死因子仪(TNF-α)表达。方法培养小鼠巨噬细胞,用不同浓度钛颗粒刺激以及刺激不同时间,通过Elisa和RT-PcR方法检测细胞MIF、TNFα的表达量以及与DNA结合的NF-κB(p65)的蛋白量。结果不同浓度钛颗粒刺激24h后,随着钛浓度增加,MIF、TNF仪蛋白量和TNFα mRNA表达逐步增加。0.1%钛颗粒刺激后,MIF、TNFα蛋白在12h开始显著性升高,24h达到高峰,36h开始下降;与DNA结合的NF-κB蛋白量在1h开始升高,3h达到高峰,6h开始下降。用PDTC阻断后,MIF、TNFα蛋白的表达明显被抑制。结论钛颗粒刺激后,先是NF-κB活化,然后上调MIF和TNF仪表达。通过这些炎症因子,假体磨损颗粒促进假体周围炎症反应,导致假体无菌性松动。objective to study the wear particles from implants up-regulating MIF and TNFα via NF-κB signal pathway. Methods The RAW264.7 were stimulated by different concentrations of titanium particles with different period of time. MIF and TNFα expression were investigated via the methods of Elisa and RT-PCR. DNA binding NF-κB ( p65 ) were investigated via the method of Elisa. Results RAW264.7 activity was inhibited by Titanium particles. After RAW264.7 were stimulated by different concentrations of titanium particles with 24h, the level of MIF, TNFα protein and TNFα mRNA increased gradually with titanium concentration increased. After RAW264.7 were stimulated by 0.1% titanium particles, the level of MIF and TNFα protein increased significantly at 12h, peaked at 24h, and began to decline at 36h; Combination of DNA and NF-κB protein began to increase at 1h, peaked at 3h, began to decline at 6h. After PDTC inhibited the activity of NF- κB, expression of MIF and TNFα protein was significantly suppressed. Conclusion After RAW264.7 is stimulated by titanium particles, expression of MIF and TNF α will increase via NF-κB activation. Wear particles from implants may promote periprosthetic inflammation, leading to aseptic loosening of prosthesis via these inflammatory factors.
