摘  要:目的研究体外条件下3-羟基丁酸(3-HB)对神经干细胞(NSCs)增殖及分化的影响。方法分离培养孕14.5d大鼠胚胎大脑皮质NSCs,分别以不同质量浓度3-HB(0.1、0.05、0.02、0.01g/L)处理培养至第3代的NSCs,以未加药组作为对照。CCK-8试剂盒检测细胞增殖、活力,免疫细胞化学染色观察NSCs的分化情况。结果与对照组相比,3-HB对NSCs的增殖、活力没有显著影响,但3-HB处理组NSCs分化为β-tubulinⅢ阳性细胞(神经元)的比例增加,而分化为GFAP阳性细胞(胶质细胞)的比例没有显著变化。结论 3-HB不影响NSCs的增殖,但可促进其向神经元方向分化。Objective To explore the effect of 3-hydroxybutyrate(3-HB) on the proliferation and differentiation of rat cortical neural stem cells(NSCs) in vitro.Methods NSCs were isolated from the cerebral cortex of rat embryos on embryonic day 14-15,and cells in passage 3 were used for all experiments.NSCs were treated with different concentrations of 3-HB(0.01,0.05,0.05 and 0.1 g/L),respectively.Untreated NSCs served as the controls.The proliferation and viability of NSCs were detected by CCK-8;the differentiation was analyzed by immunocytochemistry staining.Results There was no significant difference between 3-HB treated and non-treated NSCs regarding cell viability or proliferation.The percentage of β-tubulin Ⅲ positive cells significantly increased in 3-HB treated group,while the percentage of GFAP positive cells was similar with that in the non-treated group.Conclusion 3-HB treatment does not affect the proliferation of NSCs,but promotes the neuronal differentiation of NSCs in vitro.
