摘 要:目的建立一种简单快速测定氢溴酸加兰他敏和力可拉敏的流动注射化学发光方法。方法在碱性条件下,加兰他敏与力可拉敏对鲁米诺-铁氰化钾发光体系有明显的抑制作用,且抑制效果与药物浓度呈线性关系。据此,建立了氢溴酸加兰他敏和力可拉敏的流动注射化学发光分析方法,并采用单因素优化方法对可能的影响分析方法的因素进行了优化。结果在优化条件下,加兰他敏和力可拉敏的线性范围分别为5×10-3~10mg/L和5×10-2~10mg/L;检出限分别为0.53μg/L和2.60μg/L。结论流动注射化学发光方法可以用于氢溴酸加兰他敏和力可拉敏药物制剂和生物样品的含量分析。Objective To develop a simple and rapid flow-injection chemiluminescence(FI-CL) method for the determination of galanthamine and lycoramine.Methods In an alkaline medium,galanthamine and lycoramine can obviously inhibit the chemiluminescence intensity of the reaction of luminol-K3Fe(CN)6 system,and the relative CL intensity is proportional to the concentration of galanthamine and lycoramine.Based on this,a new FI-CL method was developed to determine galanthamine and lycoramine.And the effects of chemical and instrumental variables were optimized.Results Under the optimized conditions,the relative CL intensity was proportional to the concentration within the range of 5×10-3-10 mg/L for galanthamine and within the range of 5×10-2-10 mg/L for lycoramine.The detection limit was 0.53 μg/L for galanthamine and 2.60 μg/L for lycoramine,respectively.Conclusion The flow-injection chemiluminescence method can be applied to determine the content of galanthamine and lycoramine in pharmaceutical preparations and in serum.
