黄连配方颗粒高效液相色谱特征图谱研究

摘　　要：目的:建立黄连配方颗粒高效液相色谱(HPLC)特征图谱,为黄连配方颗粒的品种鉴别和质量控制提供参考依据。方法:色谱柱为Megres5-C18(250 mm×4.6 mm,5μm),流动相为乙腈-0.05 mol·L-1磷酸二氢钾水溶液(磷酸调节pH值为4.0)梯度洗脱,流速为1.0 mL·min-1,柱温为30℃,检测波长为345 nm,进样量为10μL。结果:由5个共有峰构成黄连配方颗粒的特征图谱,且10批样品的相似度在0.999以上。结论:HPLC法精密度、稳定性、重复性良好,可为黄连配方颗粒的鉴别和质量评价提供参考。OBJECTIVE：To establish the HPLC characteristic chromatogram of formula granule of coptidis rhizoma to provide reference for the identification and quality control of this formula granule.METHODS：The chromatograms were obtained using Megres5-C18 column（250 mm×4.6 mm,5 μm） with mobile phase consisted of acetonitrile and 0.05 mol·L-1 potassium dihydrogen phosphate aqueous solution（pH 4.0 adjusted by phosphoric acid） in gradient elution at flow rate of 1.0 mL·min-1,column temperature of 30 ℃,detection wavelength of 345 nm and sample size of 10 μL.RESULTS：The HPLC characteristic chromatogram of formula granule of Coptidis Rhizoma consisted of 5 common peaks.The similarity of 10 batches of samples was above 0.999.CONCLUSION：The HPLC method is proved to be accurate,stable and reproducible and it can provide reference for the identification and quality control of formula granule of coptidis rhizoma.
