AQP 9在羊水过少人羊膜上皮细胞的表达及地塞米松的干预效应

摘　　要：目的通过观察人羊膜上皮细胞(hAECs)水通道蛋白9(AQP 9)与羊水过少的相关性及地塞米松对AQP 9 mRNA表达的影响,探讨预防和治疗羊水异常引起的高危妊娠的新思路。方法分别获取羊水正常和羊水过少产妇的羊膜组织各15例,体外分离培养hAECs,无血清培养基中加入地塞米松按不同浓度分为5组(即0μg/mL组、10μg/mL组、20μg/mL组、40μg/mL组、100μg/mL组),培养48h后,采用实时荧光定量(real-timeRT-PCR)检测AQP9mRNA的表达。结果羊水过少较羊水正常样本AQP 9 mRNA表达量减低,其差异有统计学意义(P<0.05);经不同浓度地塞米松干预后各组,与对照组比较,AQP9 mRNA表达均增加,其差异有统计学意义(P<0.05);羊水正常样本中地塞米松40μg/mL组的AQP9mRNA的表达量最高,羊水过少样本中地塞米松20μg/mL组AQP 9 mRNA的表达量最高。结论 AQP9可能在羊水调节机制中起一定作用,地塞米松具有上调羊膜上皮细胞AQP9mRNA表达的作用。Objective To investigate the correlation between the oligohydramnios and aquaporin 9 （AQP9） and the effects of dexamethasone on AQP9 expression in human amniotic epithelial cell （hAECs） and further provide an alternative method for the prevention and treatment of high - risk preg- nancy caused by abnormal amniotic fluid. Methods hAECs were isolated from 15 cases of placenta with normal amniotic fluid and 15 cases of oligohydramnios, hAECs were cultured with serum - free medium. The normal group and oligohydramnios group were treated with different concentration of dexamethasone （ 10 μg/mL, 20 μg/mL, 40μg/mL and 100 μg/mL） for 48 h . The AQP9 mRNA expression was de- tected by real - time RT - RCR. Results The AQP9 mRNA expression in oligohydramnios group was sig- nificantly lower than that in normal group （ P 〈 0. 05 ）. Compared with the normal group, the AQP9 mRNA expression after dexamethasone treatment （ P 〈 0. 05 ）. The peak expression of AQP9 mRNA was observed in the normal group after 40μg,/mL dexamethasone treatment and in the oligohydramnios group after 20 μg/mL dexamethasone treatment. Conclusion AQP9 might play an important role in amniotie fluid regulation and dexamethasone could increase the AQP9 mRNA expression in hAECs.
