摘 要:目的:观察三七及三七花总皂苷对斑马鱼模型促血管新生作用的影响。方法:选择正常健康转基因斑马鱼及内皮细胞生长因子受体酪氨酸激酶抑制剂(VRI)诱导血管损伤转基因斑马鱼两种模型,分别予25、50、100μg/ml的三七(根)总皂苷(PNS)和三七花总皂苷(Sanqi F)干预处理,观察两者在不同浓度下对斑马鱼肠下静脉血管(SIVs)和节间血管(ISVs)生长的影响。结果:在正常健康转基因斑马鱼模型上,Sanqi F比PNS拥有更为优势的促血管新生的效果,前者出现了明显的SIVs区域出芽现象,并呈浓度依赖性;在VRI损伤模型中,三七花总皂苷也展示了更为有效的保护和恢复ISVs和SIVs血管的效果。结论:在斑马鱼模型上,三七花总皂苷和三七(根)总皂苷均有促血管新生和保护血管的效果,三七花总皂苷效果更为明显。Objective: To evaluate the angiogenic effect of total saponins extracted from root and flower of Panax Notoginseng in zebrafish model. Methods: The healthy transgenic zebrafish and transgenic zebrafish of vascular injury induced by vascular endothelial growth factor (VEGF) receptor tyrosine kinase inhibitor lI (VRI) were selected, and the zebrafish models were treated with the total saponins (25μ g,/ml, 50 μg/ml, 100 μg/ml) extracted from root and flower of Panax Notoginseng respectively. The subintestinal vessels (SIVs) and intersegmental vessels (ISVs) in zebrat2sh were assessed. Results: In healthy transgenic zebrafish, the total saponins extracted from flower of Panax Notoginseng had better angiogenic effect than the total saponins extracted from root of Panax Notoginseng with dose-dependent manner, and there were sprouting vessels from SIVs basket. In transgenic zebrafish of vascular injury induced by VRI, the total saponins extracted from flower of Panax Notoginseng had protective and recovery effects on the SIVs and ISVs. Conclusion: The total saponins extracted from root and flower of Panax Notoginseng have angiogenic effect in zebrafish model, especially the total saponins extracted from flower of Panax Notoginseng.
