摘  要:目的:以咖啡酸为指标,研究并建立大菟丝子药材的质量标准。方法:以甲醇-0.1%磷酸(75∶25)为流动相,Diamo-sil色谱柱,流速1ml.min-1,在325nm处进行咖啡酸的HPLC测定。鉴别项采用薄层色谱法(TLC)为定性鉴别方法;结果:咖啡酸在HPLC进样0.3040~2.1280μg线性关系良好(0.9999),平均加样回收率为100.03%,RSD为0.2%(n=5)。TLC能检测出咖啡酸的特征斑点;结论:该方法简单可行,可用于大菟丝子药材的质量控制。定性定量方法快速、准确、专属性强、重现性好,可用于大菟丝子药材的质量控制。Objective: Take caffeic acid as index, study and establish the quality standard of Cuscutaceae seed. Method Methanol - 0. 1% phosphoric acid (75:25 ) was used as mobile phase. Detect caffeic acid content using HPLC absorbance of 325nm and velocity of flow was 1 ml . min^-1. The column used in this method was Diamosil. Result: the optical density had a good linear relationship with caffeic acid concentration at the rang: 0. 3040 ~ 2. 1280ug (R = 0. 9999). The average recovery rate was 100. 03%, RSD 0. 2% ( n = 5 ). Characteristic spots could be found in Thin -layer chromatogram. Conclusion: the method established in this study was relatively simple and can be applied for the quality controlment of Cuscutaceae seed.
