摘 要:[目的]研究三元复合驱采油污水对水生动物基因的毒性效应。[方法]以霍甫水丝蚓(Limnodrilu hoffmeisteri)为受试生物,以浓度分别为12%、36%、24%和44%的采油污水稀释液为供试污染物,采用碱性单细胞凝胶电泳实验,以尾长(TL)、头部DNA含量(HeadDNA)和尾部DNA含量(Tail DNA)为指标检测不同浓度采油污水对水丝蚓DNA的损伤程度。[结果]三元复合驱采油污水会引起水丝蚓体细胞DNA损失,暴露组中受试生物的头部DNA含量与对照组相比均显著减少(P<0.05),尾长和尾部DNA含量均显著升高(P<0.05),随着采油污水浓度的增加,DNA损伤表现出明显的"剂量-效应"关系。[结论]该研究表明三元复合驱采油污水对水丝蚓具有基因毒性效应,可能对油田生态环境产生严重影响。[Objective] The aim was to study the genotoxicity effects of oily sewage of alkali-surfactant-polymer flooding on aquatic animals.[Method] Limnodrilu hoffmeisteri exposed to oily sewage with concentrations of 12%,36%,24% and 44%(V/V) respectively.Then the lysed cells were detected by single cell gel electrophoresis under alkaline conditions and the DNA damage of Limnodrilu hoffmeisteri cells was detected by using the tail length(TL),Head DNA content and tail DNA content as the indexes.[Result] The oily sewage of alkali-surfactant-polymer flooding could cause the damage of Limnodrilu hoffmeisteri somatopiasm DNA.The tail length and tail DNA content of all the treated groups increased significantly compared with those of control(P0.05),and the head DNA content were decreased significantly(P0.05).The DNA damage showed obvious "dose-effect" relationship along with the concentration increased of oily sewage.[Conclusion] This study showed that the oily sewage of alkali-surfactant-polymer flooding has genotoxicity effect on Limnodrilu hoffmeisteri,and may have a serious impact on the ecological environment of oil field.
