摘 要:目的:探讨高氧诱导视网膜病变(OIR)动物模型的建立及动物模型血管内皮生长因子(VEGF)基因的调节变化规律,阐明VEGFmRNA在早产儿视网膜病变模型中的表达及其意义。方法:①将10只7日龄C57BL/6J幼鼠暴露在(75±2)%浓度的高氧状态下5 d,随后在正常氧环境下5 d,作为氧诱导模型组;采用同日龄幼鼠10只作为正常对照组,空气中饲养。分别在出生后第17 d处死幼鼠。视网膜切片HE染色,观察并计数细胞核数目。②将24只7日龄幼鼠作为实验组,分为3个小组。采用同日龄的幼鼠24只作为正常对照组,同样的方法饲养,分别在出生后第12、14、17 d处死幼鼠,采用半定量逆转录-聚合酶链反应(RT-PCR)观察各组VEGFmRNA的变化。结果:①视网膜切片苏木精-伊红(HE)染色法行血管内皮细胞核计数,可见正常组仅在少数切片中见到突破内界膜的血管内皮细胞核,平均每张切片数(0.390±0.099)个;高氧组可见到较多的新生血管芽及突破内界膜的血管内皮细胞核,每张切片突破内界膜的血管内皮细胞核个数平均为(33.590±1.462),差异有统计学意义(P<0.05)。②RT-PCR结果显示,吸氧组新生小鼠第12 d OIR模型组视网膜表达VEGmRNA水平(0.295±0.038)较正常组(0.344±0.047)下降,P<0.05;第14 d OIR模型组视网膜表达VEGFmRNA水平(0.762±0.063)较正常组(0.335±0.032)增高,P<0.05;第17 d OIR模型组视网膜表达VEGFmRNA水平(0.678±0.046)较正常组(0.318±0.044)增高,P<0.05。③第14 d OIR模型组视网膜表达VEGFmRNA水平较第12 d明显升高,P<0.05;第17 d OIR模型组视网膜表达VEGFmRNA较第14 d下降,差异均有统计学意义(P<0.05)。第17 d OIR模型组视网膜表达VEGFmRNA较第12 d升高,差异均有统计学意义(P<0.05)。④正常组第12 d、第14 d、第17 d视网膜表达VEGFmRNA水平逐渐下降,经检验差异无统计学意义。结论:①新生C57BL/6J小鼠动物模型制作周期比较短,成功率比较高,是研究视网膜病变的较Objective:To investigate of the expression and change of VEGFmRNA in the model of retinopathy of prematurity(ROP) and to elucidate the expression and significance of VEGFmRNA in the model of ROP.Methods:①Put the 10 7-day-old C57BL/6J infant mice into a high oxygen conditions with the concentration being(75±2)% for 5 days and then into the normal oxygen environment for 5 days;take the infant mice of the same days(that is,7 days after the birth) to serve as the normal control group,and raise them under the air environment of(23±2) ℃.and kill the infant mice 17days respectively after their birth.and eyeball samples were parafinnembedded.The number of vascular endothelial cells were counted by retinal slices HE staining.②Put the 24 7-day-old C57BL/6J infant mice into a high oxygen conditions with the concentration being(75±2)% for 5 days and then into the normal oxygen environment for 5 days,serving as oxygen induction model group;divide the experimental group into three subgroups,and kill the infant mice 12,14 and 17 days respectively after their birth.Take the infant mice of the same days(that is,7 days after the birth) to serve as the normal control group,and raise them under the air environment of(23±2) ℃.Divide them into three subgroups,and kill the infant mice 12,14 and 17 days respectively after their birth.Adopt semi-quantitative retrovirus-Polymerase Chain Reaction(RT-PCR),and observe the changes of VEGF mRNA of each subgroup.Results:Carry out statistical processing and analysis by using software SPSS13.0,and take P≤0.05 as the standard of statistical significance of the difference.①Constriction and occlusion of the retinal blood vessels and decrease in perfusion were found under the hyperoxia condition,and dilation and proliferation of blood vessels were found under the relatively hypoxia status by retinal slice HE staining.The mean neovascular nuclei percross-section was less(0.390±0.099)in the normoxia control,while(33.590±1.462)in the hypero
