摘  要:目的探讨葡萄糖对成骨细胞株MG63增殖及细胞相关基因表达的影响。方法以不同浓度葡萄糖干预细胞株MG63(干预组),设磷酸缓冲盐溶液(PBS)为对照(对照组)。应用细胞增殖和细胞毒性检测试剂盒(CCK-8)检测细胞增殖;应用生化分析仪检测细胞内碱性磷酸酶(ALP)的含量;应用RT-PCR检测骨钙素(OCN)、骨保护素(OPG)、核因子-κB受体活化因子配体(RANKL)基因的表达。结果与对照组相比,干预组细胞增殖受抑制(P<0.01),ALP活性降低(P<0.05),OCN和OPG表达减弱(P<0.05),而RANKL的表达增强(P<0.05)。结论高糖环境可抑制成骨细胞增殖、分化和基质成熟,同时下调OPG、上调RANKL的表达,可能是糖尿病性骨质疏松的重要发病机制之一。Objective To investigate the changes of proliferation and related genes expression of human osteoblast MG63 treated by different concentrations of glucose.Methods MG63 was treated with different concentrations of glucose(group A) or by phosphate buffer saline(group B) as control.The proliferation was measured by cell counting Kit-8(CCK-8) assay,intracellular alkaline phosphatase(ALP) was detected by biochemical analyzer,and the expressions of osteocalcin(OCN),osteoprotegerin(OPG) and receptor activator of nuclear factor-κB ligand(RANKL) were examined with RT-PCR.Results Compared with group B,cell proliferation was suppressed,the expression of ALP was decreased(P0.05),and the expressions of OCN and OPG were downregulated,but RANKL was upregulated(P0.05).Conclusion High concentration of glucose could suppress the proliferation differentiation and matrix mineralization of the osteoblast,downregulate OPG expression and upregulate RANKL expression,which may be an important factor in the pathogenesis of diabetic osteoporosis.
