摘  要:目的探讨蛛网膜下腔出血(SAH)后突触体素(Syn)在大鼠脑组织中的表达变化。方法成年健康雄性SD大鼠42只,随机均分为正常(BC)组、生理盐水对照(MC)组和5个SAH模型组(D1-5组,用非肝素化动脉血注入视交叉池建模)。D1-5组大鼠分别在SAH后1、3、5、7、14d处死,取颞叶皮层、海马、小脑标本,用Western blot和免疫组化法测定Syn表达。结果与BC组和MC组比较,D1、D2、D3和D4组Syn蛋白表达明显降低(P<0.05),D5组Syn蛋白表达与BC组和MC组相仿(P>0.05)。结论 Syn的表达在SAH后第1天明显下降,其后逐渐上升,至14d时基本恢复正常。Objective To investigate the expression of synaptophysin(Syn) in brain tissues of rat model with subarachnoid hemorrhage(SAH).Methods Fourty-two SD rats were equally randomized into seven groups of NC(blank control),MC(normal saline control) and 5 SAH model groups(D1-5).SAH models were established by injection of 0.3 ml fresh arterial non-heparinized blood into the prechiasmatic cistern.On the 1st(D1),3rd(D2),5th(D3),7th(D4) and,14th(D5) day after SAH establishment,the rats were killed and brain tissus(cerebral cortex,hippocampus and cerebellum) were taken out for detecting the expressions of Syn by Western blot and immunohistochemistry.Results Compared with groups of NC and MC,the expressions of Syn in groups of D1,D2,D3,and D4 were lower(P0.05),which in group D5 were similar to those in in groups of NC.Conclusion The expression of Syn decreases significantly on the 1st day after SAH,which is increassed gradually thereafter and returns to normal two weeks later.
