摘 要:目的 :研究沙苑子提取液干预大鼠草酸钙肾结石形成的影响并探讨其作用机制。方法 :将50只大鼠随机分成5组,A组为正常对照组,B组为模型对照组,干预组(C、D、E组):在B组的基础上每天以沙苑子提取液浓度为0.05g/mL、0.1g/mL、0.15g/mL的药液分别灌胃。喂养3周检测大鼠24h尿草酸(Ox),钙(Ca2+)浓度;血尿素氮(BUN),肌酐(Cr)含量;肾组织超氧歧化酶(SOD)的活性,丙二醛(MDA)含量;观察大鼠肾组织草酸钙结晶沉积情况;免疫组化检测大鼠肾组织骨桥蛋白(OPN)的表达水平。结果:干预组大鼠24h尿Ox、Ca2+浓度,血清Cr较B组明显降低(P<0.05)。肾组织SOD的活力,C、D、E组较B组显著增高(P<0.05);MDA含量,C、D、E组较B组明显降低(P<0.05)。OPN在各组中均有表达,A组中仅能检测到OPN的微弱表达,B组OPN表达则明显增强;干预组低于B组。结论:沙苑子能够减少肾脏内草酸钙晶体沉积,其机制可能通过降低OPN表达,减轻肾损伤,抑制大鼠草酸钙结石形成。Objective: To investigate the effect of Semen Astragali Complanali extract on the formation of renal calcium oxalate stone and to explore the possible mechanism. Methods: Fifty adult Sprague Dawley rats were randomized into 5 groups, group A(normal control), group B(model control), and group C, D, E(model rats treated with 0.05, 0.1, and 0.15g/mL of Semen Astragali Complanali extract solution by intragastric administration respectively). After feeding for 3 weeks, we detected the contents of 24-h urinary oxalic acid(Ox), urinary calcium, blood urea nitrogen(BUN) and serum creatinine (Cr), observed the renal superoxide dismutase(SOD)activity and renal malondialdehyde(MDA) content as well as the deposition of calcium oxalate crystal, and examined the renal osteopontin(OPN) expression level by immunohistochemistry. Results: The urinary 24-h Ox and calcium concentration and serum Cr were obviously lower, renal SOD activity was higher, and renal MDA content was lower in group C, D and E than those in group B(P 〈 0.05). OPN expression was weak in group A, strong in group B and then became weak in group C, D and E. Conclusion: Semen Astragaii Complanali extract exerts an effect on inhibiting the formation of renal calcium oxalate stone through relieving the deposition of calcium oxalate crystal and reducing OPN expression level. They may reduce the Renal injury and inhibit the expression of OPN, thus inhibit calcium oxalate crys- tal deposition in rat kidney.
