摘 要:目的分析系统性红斑狼疮(SLE)患者外周血中调节性T细胞(Treg)表达情况,初步探讨其在SLE患者细胞免疫失调中的意义。方法采用流式细胞术检测50例SLE患者和31例健康人对照组外周血中CD4+CD25+CD127low/-Treg细胞百分率,并分析其与CD4+CD25+和CD4+CD25high细胞的相关性,以及各种Treg细胞与SLE患者及SLE疾病活动性指数(SLEDAI)的相关性。结果 SLE活动期外周血CD4+CD25high细胞百分数为(11.65±5.53)%,显著高于SLE稳定期的(8.52±3.15)%和健康人对照组的(6.85±1.63)%,F=23.287,P<0.01。SLE患者Treg细胞与CD4+CD25high和CD4+CD25+细胞百分数均呈正相关(r分别为0.819,0.711,P均<0.05),SLEDAI与CD4+CD25+CD127low/-、CD4+CD25+、CD4+CD25high细胞水平呈正相关(r分别为0.410,0.394,0.348,P均<0.05)。结论 SLE活动期外周血中Treg细胞表达增加,且与SLEDAI相关,SLE患者免疫异常可能与Treg细胞的异常水平有关。Objective To analyze the expression of regulatory T cells(Treg) in peripheral blood of patients with systemic lupus erythematosus(SLE),and explore the role of Treg in cell immunity disorder of SLE.Methods The percentages of Treg with CD4+CD25+CD127low/-in preipheral blood from 50 patients with SLE and 31 healthy controls were determined by flow cytometry.The correlations between CD4+CD25+CD127low/-T cells and CD4+CD25+ or CD4+CD25high T cells,and between the expressions of all kinds of Treg cells and SLE disease activity index(SLEDAI) were analyzed.Results The percentage of CD4+CD25high T cells in patients with active SLE [(11.65±5.53)%] was significantly higher than that of patients with stable SLE [(8.52±3.15)%] and of the healthy control [(6.85±1.63)%](F=23.287,P0.01).The percentage of Treg cells in SLE patients was positively correlated with that of CD4+CD25high T cells(r=0.819,P0.05),and of CD4+CD25+ T cells(r=0.711,P0.05).Moreover,there were positive correlations between SLEDAI and CD4+CD25+CD127low/-,CD4+CD25+ and CD4+CD25high T cells repectively for r=0.410,0.394 and 0.348(P0.05).Conclusion The percentage of Treg cells in peripheral blood of patients with active SLE was increased,and positively correlated with SLEDAI,indicating that the immunity disorder of SLE may be related to the abnormal expression of Treg cells.
