摘 要:目的研究羊水内活细胞形态及贴壁率的差异,探讨快速贴壁细胞与神经管缺陷(NTD)之间的关系,检测羊水中神经源性细胞阳性表达率,探讨其与神经管畸形之间的关系。方法选取在甘肃省妇幼保健院门诊抽取羊水做产前诊断(后荧光原位杂交证实为正常胎儿)和已被证实为神经管畸形的孕妇羊水做常规羊水培养,观察羊水正常组(对照组)与NTD组(实验组)细胞在形态、贴壁时间的差异,并选用神经元特异性标志物,抗微管相关蛋白anti—MAP2、抗神经元核单克隆抗体(anti—NeuN),通过免疫荧光法鉴定对照组(15例)与实验组(11例)神经元特异标志物的阳性表达率。结果实验组细胞的活细胞贴壁率明显高于对照组,差异有统计学意义(t=8.909,P=0.000)。免疫荧光法结果显示羊水正常组(对照组)与空白组(未干预)两种神经源性细胞标志物表达均为阴性,NTD组(实验组)抗神经元核单克隆抗体(anti—NeuN)表达阳性,抗微管相关蛋白anti—MAP2表达阴性,对照组与实验组组间阳性率差异有统计学意义(X2=7.717,P=0.021)。结论检测羊水中神经源性细胞可辅助诊断神经管畸形。Objective To explore the relationship between rapidly adhering cell (RAC) and neural tuber defect (NTD) by studying the differences in morphology and adhesion rate of live cells, and to explore the correlation between positive expression rate of neurogenic cells in amniotic fluid and NTD. Methods The amniotic fluid from pregnant women who did prenatal diagnosis ( confirmed with normal fetus by FISH) in outpatient of Gansu Provincial Maternity and Child-care Hospital and that from women confirmed with NTD were cultured. The differences in morphology and adhesion rate between normal amniotic fluid group (control group) and NTD group (experimental group) were observed. Then the neuron-specific marker of anti-MAP2 and anti-NeuN were selected to identify the positive expression rates of neuron-specific markers in control group (n = 15 ) and experimental group (n = 11 ) through immunofluorescenee. Results Adhesion rate of live cells of the experimental group was significantly higher than that of control group and the difference was significant ( t = 8. 909, P = 0.000). Immunofluorescence results revealed negative expr^ession of neurogenic cells in the control group and blank group. The expression of anti-NeuN was positive and expression of anti-MAP2 was negative in NTD group. The differences were statistically significant between control group and experimental group(x2 = 7. 717, P = 0. 021 ). Conclusion Detection of the neurogenic cells in amniotic fluid can assist the diagnosis of NTD.
