摘  要:为了优化高山杜鹃茎段离体组织培养体系及其工厂化繁育体系,对10份不同基因型高山杜鹃离体培养与植株再生差异进行了研究。结果表明,供试高山杜鹃茎段外植体在WPM培养基上均能诱导出芽、增殖和生根。在诱导萌发阶段,添加0.5~1.0 mg/L的ZT萌发效果较好,其中有7份材料在0.5 mg/L ZT时获得最佳萌发诱导。在增殖阶段,最佳增殖激素配比为ZT 0.5~0.7 mg/L+NAA 1.0 mg/L,其中6份材料在ZT 0.5 mg/L+NAA 1.0 mg/L激素配比下增殖达6~9倍。生根阶段,培养基最佳激素配比则为NAA 0.25mg/L+IBA 0.25 mg/L。并且试管外生根效果好于试管内生根,平均成活率为80.95%,而试管内平均成活率仅为57.94%。在最佳激素配比下供试所有高山杜鹃采用试管外生根成活率均在98%以上,并且试管外生根成本低,操作简便快速,适用于工厂化生产。To optimize the in vitro culture system and factory propagation system from stem fragments, the differences of in vitro regeneration and culture among ten genotypes of Rhododendron lapponicum were in- vestigated. The results indicated that the explants of stem fragments from all Rhododendron lapponicum geno- types could be induced to germinate, proliferate and take root on the WPM medium. During the process of in- ducing adventitious buds, the germination efficiency was better by adding 0. 5 - 1.0 mg/L zeatin, in which, seven genotypes got the best germination at 0. 5 mg/L zeatin. At the multiplication stage, 0. 5 ± 0. 7 mg/L ZT + 1.0 mg/L NAA was the best, and the multiplication rate of six genotypes treated by ZT 0. 5 mg/L + NAA 1.0 mg/L was about 6 -9 times higher than that without any hormone treatment. At the rooting stage, the best hormone ratio was NAA 0. 25 mg/L + IBA 0. 25 mg/L, and the rooting out of tube with the survival rate of 80. 95% was better than that in tube with the survival rate of 57.94%. Under the hormone treatment with the best ratio, the survival rate of all genotypes was over 98%. Meanwhile, the rooting out of tube cost lower and could be operated more conveniently, so it was suitable for factory propagation.
