湿地松丛生芽离体保存技术的研究

摘  要:为了有效保存和利用现有的抗病湿地松优良品种资源,本文对湿地松优良种质资源的离体保存技术进行了研究。结果表明:温度、基本培养基、蔗糖浓度及培养基中添加抑制剂等对湿地松丛生芽低温保存后生活力有显著影响。14℃环境下丛生芽保存10个月后,成活率可达到96.8%,生长恢复测定时的增殖系数高达6.2,生根率高达100%。1/2GD+甘露醇5mg/L+蔗糖10g/L或1/3GD+甘露醇10mg/L+蔗糖5g/L的组合比较适合于湿地松丛生芽低温保存。In vitro conservation technique of slash pine ( Pinus elliottii) was studied to effectively conserve the germplasm resources , which have good quality and can strongly resist to the brown spot needle blight. The viability of axillary buds was found to be influenced by temperature, basal media concentration, sucrose concentration as well as the retardant concentration. After 10 months storage in 14℃, survival rate and rooting rates of axillary buds were up to 96. 8% and 100% , respectively. The proliferation multiple was 6.2. The optimal conservation condition was half strength of modified Gresshoff and Doy (GD) medium supplemented with 5 mg/L mannitol and 10 g/L sucrose or one third strength of modified GD medium supplemented with 10mg/L mannitol and 5 g/L sucrose.
