摘 要:探讨了光合细菌3号菌株产类胡萝卜素培养基的优化以及类胡萝卜素的提取.首先对3号菌株的60%蔗糖悬液进行全波长扫描,确定了3号菌株在457nm波长下出现最大吸收峰,因此用457nm波长下的OD值作为类胡萝卜素相对含量高低的指标.通过改变培养基碳源、氮源和盐度确定最佳的碳源、氮源和盐度,得出最优碳源为蔗糖,最优氮源为硝酸铵,最适盐度为0.1‰.利用最佳培养基培养3号菌株,用皂化法分离纯化类胡萝卜素,得类胡萝卜素纯品,提取率为30.06mg.g-1.The paper mainly discusses the medium optimization of photosynthetic bacteria strain 3 high yield of caroteholds and the extraction of carotenoids. The No. 3 strain, 60% sucrose levitation liquid is used for the whole wavelength scanning to determine the maximum absorption peak of strain 3. The result is 457 nm, so it is considered as the carotenoid relative content discretion indicator. By changing the culture medium carbon sources, nitrogen sources and salinity to determine the best source of carbon, nitrogen and salinity, and they are sucrose, ammonium nitrate and 0.1‰. Use the best culture medium to train the photosynthetic bacteria strain 3, then use saponification method to separate and purify the carotenoids. The carotenoids are tasted, and the extraction rate is 30.06 mg · g^-1.
