摘 要:目的:建立大鼠血浆中异硫氰酸烯丙酯含量测定的UPLC法,并用该法研究大鼠静脉注射异硫氰酸烯丙酯后的药动学。方法:大鼠血浆样品,采用乙腈沉淀蛋白分析。色谱柱:BEH C18色谱柱(2.1 mm×100 mm,1.7μm);流动相:甲醇-水(60∶40);流速0.2 mL·min-1;柱温30℃;紫外检测波长247 nm;大鼠静脉注射异硫氰酸烯丙酯20 mg·kg-1后,于不同时间点取血测定血浆中药物浓度并估算其药动学参数。结果:异硫氰酸烯丙酯在0.156~40.0μg·mL-1范围内,其线性关系良好,r=0.998 9,定量下限为0.156μg·mL-1,提取回收率为91.34%。大鼠静脉注射20 mg·kg-1异硫氰酸烯丙酯后其消除半衰期为77.36 min。结论:本方法灵敏度高,操作简便、快速、准确,可用于大鼠血浆中异硫氰酸烯丙酯的测定及药动学研究。OBJECTIVE To establish a simple assay method for allyl i^othiocyanate in rat plasma by ultra performance hqmd chromatography (UPLC), and investigate its pharmacokinetics in rats with intravenous injection. METItODS Plasma samples were treated with acetonitrile to deposit proteins. BEH C18 column (2. 1 mm×× 100 mm, 1.7 μm) was used. The chromatographic conditions were as follows: the mobile phase was methanol distilled water (60:40) ; the flow rate was 0. 2 mL.min^-1 the column temperature was at 30 ℃ and the UV detection wavelength was set at 247 nm Rat blood samples were collected at different intervals after intravenous injection of a single dose (20 mg. kg^-1 ) of allyl isothioeyanate and the concentration of allyl isothiocyanate in rat plasma were determined by UPLC method for estimating pharmacokinetic parameters. RFNULTS There was a linear relationship of the concentration of allyl isothiocyanate within the range of 0. 156 to 40. 0μg,mL-1 (r= 0. 998 9). The recovery rate was 91.34%. The lower limit of quantitation of this method was 0. 156 μg.mL^-1. The elimination half life was 77. 36 min in rats with intravenous injection of 20 mg.kg ^-1 allylisothiocyanate. CONCLUSION The method is high sensitivity, simplicity, fast, accuracy and reliance for the determination of allylisothiocyanate in rat plasma and can be used for the study of pharmaeokineties of allyl isothiocyanate.
