摘 要:目的研究α-细辛醚对癫痫大鼠的海马钙稳态的影响。方法大鼠癫痫持续状态(SE)后,观察α-细辛醚对海马游离Ca2+浓度、游离钙调蛋白(CaM)平均通道荧光值、总CaM及钙调蛋白依赖性蛋白激酶(CaMK)IIa-mRNA表达的变化的影响。结果模型组SE后,各时相海马游离Ca2+显著增高(P<0.05);CaM平均通道荧光值显著降低(P<0.05);SE后12~72 h,游离CaM表达较正常组明显增强(P<0.05);CaMK IIa-mRNA表达显著降低(P<0.05)。与模型组相比,α-细辛醚组游离Ca2+浓度显著降低,游离CaM平均通道荧光值明显增加,总CaM表达显著降低,CaMK IIa-mRNA表达明显增强(P<0.05)。结论α-细辛醚可下调SE后海马游离Ca2+浓度及结合CaM表达,并上调CaMK IIa-mRNA表达。Objective To observe the effects of α - asarone on hippo- campus calcium homeostasis in in epilepsy rats. Methods Rat status epilepticus (SE) model was induced by Lithium -Pilocarpine. The effects of α - asarone on the content of intracellular free Ca2+ , the average passage fluorescence of relative activity of calmodulin (CaM) and the expression of the CaM and Ca2+/CaM -dependent kinase IIa (CamK IIa) in hippocampus of post -SE rats were observed. Results The intracellular free Ca2+ content was significantly increased in the hippoeampus of epilepsy rats (P 〈 0. 05). The mean channel fluorescence of intra- cellular free CaM was significantly decreased at different time post -SE (P 〈 0. 05 ). At 12 - 72 h post - SE, the CaM expression in model group was significantly higher than those in normal group, and still higher than in normal group at one week post - SE. The expression of CaMK IIa - mRNA was significantly decreased at different time in the hippoeampus of post - SE rats ( P 〈 0. 05 ) , and still lower than in normal group at one week post - SE. The content of intracellular free Ca2 + and the expression of CaM in α - asarone group were significantly lower than those in model group at different time in the hippoeampus of post - SE rats (P 〈 0.05). The mean channel fluorescence of intraeellular free CaM and the expression of CaMK IIa - mRNA in α - asarone group were significantly higher than those in model group (P 〈 0. 05 ) . Conclusion α - asarone could significantly downregulated the intracellular free Ca2+ content and the ex- pression of CaM and upregulated the expression of CamK IIa in the hippocampus of post - SE.
